Summary: Streptomyces rimosus R6 normally carries a giant linear
plasmid (GLP) of about 390 kb. During a strain improvement programme using
a high producer, derivatives were obtained with increased oxytetracycline
(OTC) production and resistance. Most of them had DNA rearrangements in the
chromosome, except one strain (MV17) that had a normal chromosomal pattern,
but an altered GLP. Closer analysis showed that MV17 possessed a GLP' of
about 1 Mb in size that was derived from parental GLP and carried the
OTC-region. The increased production and resistance of MV17 is presumably
due to the increased copy number of the OTC-cluster, which is present both
on the chromosome and on the GLP'. Possible mechanisms for the genetic
instability of OTC production in MV17 and for the formation of GLP'
plasmids are discussed.

Summary: 'Strain degeneration' is wide-spread in industrial
Streptomyces strains and describes a variety of spontaneous changes that
result deleterious phenotypes especially for antibiotic yield. Genetic
instability (i.e. the occurrence of spontaneous mutations with a high
frequency) is common in Streptomyces and has been studied in several model
systems, where a pattern of large-scale DNA deletions and amplifications
has been established. However, little work has been performed with
high-yielding industrial strains to define the relationship between 'strain
degeneration' and genetic instability. S. rimosus R6 strains that have been
selected for high oxytetracycline production show genetic instability. Two
classes of mutants were defined with reduced or no antibiotic production.
The Class II mutants carried large deletions including the oxytetracycline
gene cluster, which resulted in the loss of both the production and
resistance genes. Although this proves that the genes lie in an unstable
chromosomal region, the fact that the mutants are sensitive to
oxytetracycline makes them unimportant for 'strain degeneration'. In
contrast, the commonest low-producing mutants (Class I) remained resistant
to the antibiotic and outgrow the parent strain in competition experiments.
This suggests that the Class I mutants, which did not show any detectable
DNA rearrangements, are responsible for 'strain degeneration'. A hypothesis
to explain Class I mutants will be discussed.

Summary: Recent studies imply that a linear chromosome may be
characteristic of the Streptomyces genus. S. rimosus R6, the
oxytetracycline (OTC) producer, carries a linear plasmid pPZG101 that can
interact with the bacterial chromosome to form integrants and
plasmid-primes. Restriction analysis of one integrant (MV25) showed that
pPZG101 sequence had been integrated into the chromosome together with its
linear end(s). Another strain (MV17) carried a linear plasmid of increased
size of about 1 Mb (pPZG103). It was shown that OTC-genes were carried on
the pPZG103, indicating that the plasmid was pPZG101'-OTC. Restriction
mapping of pPZG103 demonstrated that one plasmid end was derived from the
parental pPZG101, while the other end was linear chromosomal end. The
linearity of S. rimosus R6 chromosomal DNA was further supported by the
development of the chromosomal physical map.
