Summary: Three herbicides (SAN 9789, SAN 9785 and amitrole) were
used tostudy the biosynthesis of carotenoids and structural changes
inchromoplasts of Calceolaria flowers. All three herbicidesaffected the
biosynthesis of total carotenoids, especially oflutein. SAN 9789
considerably inhibited the normal formation ofchromoplast membranes.
Amitrole moderately influenced thedevelopment of these structures, while
SAN 9785 did not cause anyconspicuous fine structural modifications in the
chromoplasts.

Summary: The changes in number, size, distribution and DNA content
ofplastid nucleoids in wild- and mutant-type leaves of an aureavariety of
privet were studied. Wild-type leaves did not showchanges induced by
different light intensities. Mutant leavesresponded to changes in light
conditions by changes in colour.During yellowing of regreened aurea leaves
and bleaching ofyellow aurea leaves the number of plastid nucleoids
decreasedwith concomitant increase of their volume in such a fashion
thatthe total volume of nucleoids of the DNA content per plastidremained
constant.

Summary: The effects of the bleaching herbicide amitrole on the
carotenoidcomposition and ultrastructure of tulip tree chromoplasts
werestudied. Amitrole inhibited the accumulation of beta-carotene andof an
unidentified carotene. The biosynthesis of xanthophylls wassimilar to that
in the control. Amitrole caused the appearance oflong closely packed
unbranched tubules in some zones of thereticulum. At the last stages of
flower development in thechromoplasts treated only plastoglobules with
small carotenoidcrystaloids remained.

Summary: The fine structure of plastids and carotenoid content of
the pepper fruit were studied during ripening. Red ripe fruits contained
spidle-shaped chromoplasts which originated from leuco-chloroplasts. The
bleaching herbicides amitrole and SAN 9789 inhibited the biosynthesis of
total carotenoids and the development of typical spidle-shaped
chromoplasts. Both herbicides prevented the formation of tubules and
increased the accumulation of plastoglobules.

Summary: The leaves of Sophora japonica remain green througt the
autum, then fall off late in November or beginning of December without
change in their colour. The ultrastructure of the chloroplasts was normal
during that time. The gradual increase of plastoglobules and the presence
of some interthylakoidal inclusions showed that senescence was in progress.

Summary: Two horse-radish crown-gall tissue lines, one unorganized
and the other teratoma producing were compared with the control leaf in
ultrastructure and electrophoretic protein patterns. Tumour cells were
similar to unspecialized parenchyma cells. Large lobe nuclei were observed
in both types of transformed cells. Leaf specific polypeptides of about
22-, 21-, 20 and 18 kDa disappeared in tumour extracts. The polypeptide of
about 85 kDa was present only in transformed tissue.

Summary: The ultrastructure and pigment composition of aurea maple
treeleaf chloroplasts were studied. Chloroplasts of aurea type leavesare
normally developed in low light ilumination. High lightconditions lead to
gross ultrastructural damage of thylakoidmembranes, reduction of
chlorophyll content, loss of somecarotenoids and degradation of the protein
component of LHCIIapoprotein. It can be concluded that these are
consequences ofdisturbed adaptation to high light conditions.

Summary: When potato tuber slices were exposed to light they become
greenin a few days. During greening the amyloplasts were transformedinto
chloro-amyloplasts with normal thylakoid system. Besidesthese, small
chloroplasts probably transformed from leucoplasts,were also visible. Apart
from ultrastructural changes plastidtransformation includes changes in
pigment and protein content.

Summary: Crown gall tumours were induced on horse-radish
(Armoracialapathifolia Gilib.) leaf fragments by Agrobacterium
tumefaciensB6S3. Two different tumour lines, one unorganized and the
otherorganized teratoma were compared. The obtained SDS-proteinbanding
patters were rather similar. Peroxidase activity washigher in the
teratogenous tumour than in the unorganized tumourtissue. The
ultrastructural localization of peroxidase revealeda DAB reaction product
in the tonoplast surrounding the largevacuole of tumour cells.

Summary: The roots of some vascular plants when held in light become
green. The chloroplasts appeared first in the oldest root zones in the
innermost parenchyma cell layers. The greening then spread peripherally and
into younger root segments. The number of photosynthetically active
chloroplasts increased during greening.

Summary: Phenotypic characterization of the Arabidopsis thaliana
mutant BEL1 revealed numerous morphological abnormalities of BEL1 ovules,
including the absence of the inner integuments, homeotic transformation of
the outer integuments into carpel-like structures, and incomplete
development of the female gametophytes. The homeotic transformation of
outer integuments into carpelloid structure in BEL1 has been correlated
with abnormal expression of class C gene expression in the Arabidopsis
ovule. The search for genes homologous to BEL1 resulted in identification
of three new homeobox genes, ABH1, ABH2, and ABH3. Together with BEL1,
these three genes define a novel homeobox gene subfamily in Arabidopsis.

Summary: The process of amyloplast-chloroplast transformation
underspecial light conditions was studied by light and electronmicroscopy.
Already one day after the beginning of the experimentit was possible to
detect primary red fluorescence ofchlorophyll. Electron microscope analysis
showed ultrastructuralchanges of amyloplasts which include thylakoid
formation andtheir aggregation into grana. These structural changes
werefollowed by increase of chlorophyll content and changes inprotein
picture.

Summary: The plastid ultrastructure, pigment composition, protein
andptDNA content of aurea leaves were examined. Plastids from
greenaurea-type leaves are in all studied characteristics similar tothe
wild-type ones. Plastids from yellow and bleached aurea-typeleaves have
severely altered ultrastructure. HPLC showed thattheir xanthophyll pool is
deficient and consists mainly ofviolaxanthin. Analysis of thylakoid
proteins by SDS-PAGE revealedlack of the major antenna component.
Microphotometry of DAPIstained ptDNA showed that aurea plastids have in all
stages ofplastid development 20% less DNA.
