Title: Freshly frozen subsequently AMeX processed breast
carcinoma tissue-a new possibility for immunohistochemical detection of
c-myc oncoprotein

Summary: Normally, oncoprotein p62 c-myc is localized in the cell
nucleus.In freshly frozen sections, normal immunohistochemicallocalization
of this antigen is preserved but some morfologicaldetails have been
hampered by destruction of cell and tissuearchitecture during
cryosectioning. Previously we have shown thatin paraffin embedded sections
the commonly observed cytoplasmicstaining for p62 c-myc resulted from
inadequate fixation. Wefound that a more adequate fixation procedure
included treatmentwith cold acetone (- 20 o C) hold by methyl benzoate and
xylene,(AMeX). We wished to eliminate artifacts of cryosectioning
andfixation in formalin. In this study we evaluated the AMeXprocedure in
immunohistochemical localization of p62 c-myc ininfiltrating ductal berast
carcinoma tissue freshly frozen, AMeXtreated and the embedded in paraffin
(FF-AMeX).To delineate thedisribution and intenity of staining for p-62
c-myc in tumourcells, we used the monoclonal antibody 155-11C7 raised
agains asyntetic myc peptide. We assesed the intensity of immunostainingand
percentage of positive tumour cells and observed more intensestainig of p62
c-myc in a freshly frozen section than in FF-AMeXsections, bat tissue and
cells architecture were much betterpreserved in FF-AMeX. Our results show
that the AMeX procedurein previously frozen tissues resulted in
reproducible nuclearimmunostaining for p62 c-myc. This finding is importent
becausetissues often capt frozen for steroid receptor analysis can bealso
used for immunohistochemical localization of oncoproteins.
