Summary: A series of Leu-enkephalin-related glycoconjugates with an ester-, ether, or amide-type linkage were synthesized and evaluated for antiviral activity against HIV-1 in a cell-culture system using peripheral blood lymphocytes. All tested glycoconjugates exhibited a certain antiviral activity which was significantly higher than the activity of the parent peptide compound itself. These results indicate that synthetic glycoconjugates of opioid peptides are good candidates for the development of anti-HIV agents.

Summary: The preparation and characterization of the essential components to be used in the radioimmunoassay of peptidoglycan monomer (PGM) is described. 14-C-Labeled-PGM-bovine serum albumin conjugate was prepared which elicited anti-PGM response in rabbits. Anti-PGM antibodies and 125-I-labelled Boc-Tyr-PGM were used as essential components in competitive radioimmunoassay for PGM determination in mammalian and human sera and plasma, respectively.

Summary: Three N-glycoconjugates of the general formula H-Tyr-Gly-Gly-Phe-Leu-NH-R (R= carbohydrate residue) were synthesized in order to determine the influence of some carbohydrate molecules on the biological activity, conformation, and stability of the opioid pentapeptide Leu-enkephalin. For the preparation of this compounds different methods of peptide synthesis were investigated. In comparison with Leu-enkephalin, all three N-glycoconjugates showed higher potency in the guinea pig ileum assay and lower potency in the mouse vas deference assay, indicating a decrease in delta opioid receptor selectivity.

Summary: The complexes of Fe(III) with small molecules (glycopeptides, amino sugars) related to peptidoglycan monomer (PGM) structure were prepared and characterized by chemical and physicochemical methods. A simple method for preparation using separation of there action products on a molecular sieve was introduced. Using this method, monomeric complexes of small molecular weight were obtained. The likely structures were proposed on the basis of analytical results.

Summary: Synthetic leucine-enkephalin glucoconjugates were studied by several one- or two-dimensional NMR methods. Using ROESY spectra, it was possible to detect several inter-and intra-residual nuclear Overhauser effects. The results do not support the existence of a single folded conformation, indicating that the structures of glycoconjugates studied are not completely random.

Summary: Synthesis and conformational analysis of muramic acid delta-lactam structures, characteristic of bacterial spore peptidoglycan, is described. NMR spectroscopy indicated that the glucopyranose ring in lactam derivatives adopts B(O,3)conformation. On cleavage of 1,6-anhydro ring, the glucopyranose ring adopts 4C1 conformation. X-ray analysis confirmed the proposed structures.

Summary: 14-C-Labelled chlorothalonil (2) was prepared by catalytic chlorination of 14-C-UL-dicyanobenzene on activated carbon, in a tubular reactor at acontrolled temperature and in a stream of chlorine. On purification by column chromatography on silicagel, 99.6% pure 14-C-UL-2 was obtained.

Summary: A novel type of compound that contains a peptide specific for bacterial peptidoglycans (immunomodulatory activity) and an adamantyl residue (antiviral activity) was synthesized. Thus, L-Ala-D-gama-Gln was linked to DL-Gly(Ada) yielding two diastereoisomers which were easily separated by column chromatography and characterized. The cytotoxic effect and antiviral activity of the novel adamantylpeptides were studied on MDCK cells in vitro. Both diastreoisomers exhibited low cytotoxicity and reduced the infectivity of the tested influenza A virus strains, but to a different extent on different strains.

Summary: A series of glycoconjugates, in which Met-enkephalin or D-Ala,Met-enkephalin have been linked through an ester bond to the HO-6 of various D-glycopyranose moiety, were synthesized by classical solution methods. The biological activities of these compounds were determined on selective pharmacological preparations: guinea pig ileum and mouse vas deferens for opioid activity, and two mouse cell lines, fibroblast L929 and melanoma B 16BL6, to study the influence on growth processes. The results reported in this study demonstrate the differential effect of the carbohydrate part in enkephalin-related glycoconjugates on receptor recognition.

Summary: A 14-C labelled low molecular weight immunomodulator, peptidoglycan monomer (14-C-PGM), was injected intravenously in mice. Shortly after injection, 14-C-PGM was very rapidly excreted from the organism, so that 1 h following administration, 80% of the radioactivity was found in the urine. Six hours after injection, equal quantities were found in the intestines, liver and blood (0.5%), slightly less in the kidneys, lungs and spleen (0.2-0.3%) and the least quantity in the brain (0.04%). However, the dynamics of the retention in the organs was evidently different. In the kidneys, lungs and spleen, radioactivity steadily decreased, in the liver remained the same, whereas, in the intestine and brain, PGM seemed to accumulate.

Summary: Acetylated D-glucopyranosyl esters of enkephalin were prepared by two different fragment condensation procedures involving direct participation of imidazole in the ester linkage formation. By both methods anomeric mixtures of D-glucosyl esters were obtained and resolved by column chromatography. Depending on coupling conditions, racemization of either the C-terminal or the penultimate amino acid residue of the enkephalin molecule occurred. The glucoconjugates with inverted stereochemistry were quantitated and separated from the main product by reversed-phase high-performance liquid chromatography. the opioid agonist potencies of the synthetized glucopyranosyl esters of enkephalins on electrically stimulated guinea pig ileum and mouse vas deferens preparations were determined in comparison with Leu-enkephalin.

Summary: Synthesis of the dolastatin 10 (1) dipeptid segment N-(tert-butoxycarbonyl)-dolaproinyl-dolaphenine (2, Boc-Dap-Doe) and four chiral isomers (3-5, 13) has been summarized. Formation of the amide bond was readilly accomplished employing diethyl cyanophosphonate. The stereochemical assignments for Boc-Dap-Doe (2) and Boc-Dap-(6R)-Doe (3) were confirmed by X-ray crystal structure analyses.

Summary: The chromatographic behaviour of leucine-enkephalin-related glycoconjugates with an ester-, ether- and amide-type of linkage was investigated by reversed-phase high-performance liquid chromatography using trifluoracetic acid as ion-pairing agent and methanol as modifier of the aqueous phase. The results show that the position and the type of sugar-peptide linkage, the type of sugar moiety introduced and the degree of carbohydrate protection contribute to the overall retention of the glycopeptides studied.

Summary: Reversed-phase high-performance liquid chromatographic elution data for methionine-enkephalin-related glycoconjugates were analysed as a function of the identity and position of the sugar-peptide linkage. It was shown that binding to the column could correlate with the degree of sugar moiety protection. Replacement of either the phenylalanine or methionine residue in the peptide backbone of the glycoconjugates with its D-enantiomer leads to a considerably stronger retention on a reversed-phase column. The dependence of retention times on the methanol concentration in the mobile phase suggested that, under the conditions studied, there are different retention mechanisms for glycopeptides containing unprotected sugar moieties in the molecule.

Summary: Small enkephalin-related peptides containing a 1-adamantanamine moiety coupled through an amide linkage at the C-terminus were synthesized. Several of the compounds showed high mu opioid activity and mu receptor selectivity. The new adamantanamine derivatives were also examined for antiviral activity against HIV-1 in a cell culture system. Some of them inhibited syncytia formation even when the antigen assay gave evidence for viral replication.

Summary: The title compound, a precursor in the synthesis of other analogues of Amadori's type, is prepared and characterized by X-ray structure analysis and NMR spectroscopy. The molecule is flexible even in solid state; at 100 K two orientations of the benzyloxy group are present in the unit cell. The beta-D-fructopyranose moiety blocked with two dioxolane rings is the less flexible part of the molecule; in the solid state and in DMSO-d6 it is in the skew conformation.

Summary: Seven solvents considered appropriate for use with relatively sensitive biosynthetic products undergoing separation by multilayer coil planet centrifuge high speed countercurrent distribution (HSCCD) have been evaluated in a new and systematic procedure for optimal solvent selection. The new mwthod was illustrated by a complete HSCCD separation of five closely related dipeptides.
