Summary: New biologically active compounds or their complexes
belonging to the class of monosaccharides, oligosaccharides, peptides and
glycopeptides were synthesized, or isolated from biological sources, and
their physico-chemical and biological properties examined. The following
lines of investigation were pursued: 1) synthesis and bioevaluation of
glycoconjugates in which enkephalins, endogenous opioid peptides, or some
other biologically active molecules, were coupled to free or protected
carbohydrates, 2) synthesis and conformational analysis of muramic acid
delta-lactam structures characteristic of bacterial spore peptidoglycan
units, 3) pharmacokinetics of 14-C labeled immunomodulator peptidoglycan
monomer after intravenous administration in mice.
Research goals: The objective of the proposed research is synthesis
(or isolation from biological materials) of well-defined biologically
active compounds belonging to the class of sugars, peptides and
glycopeptides, which will provide information about structure and
reactivity relationships. Information of this kind should be relevant to
better understanding of some natural events or for the design of
carbohydrate-peptide based drugs. New synthetic approaches to carbohydrate
derivatives of enkephalins, endogenous opioid pentapeptides, as a new
class of compounds of low toxicity with probable immunomodulatory,
anti-viral or anti-tumor activity will be studied. In order to gain an
insight into biological events leading from bacterium to spore
transformation, muramic acid delta-lactam related structures will be
synthesized and their conformation studied. To explain the
immunomodulatory activity of peptidoglycan monomer, bacterial cell-wall
constituent, pharmacokinetics in mice after intravenous administration
will be studied.
