Summary: 1. Erythrocytes, polymorphonuclears, monocytes and
lymphocytes isolated from human peripheral blood, were shown to possess in
their cytosols, granules and microsomal fractions, aminopeptidases capable
of hydrolysing arginyl-, leucyl-, methionyl-, phenylalanyl- and
alanyl-2-naphthylamide.
2. In different cell compartments enzymes of different pI were
responsible for these activities.
3. Chloride activated arginine aminopeptidase, broad specificity
aminopeptidase and dipeptidyl peptidase III were found in cytosols of all
examined cells.
4. In granules at least two aminopeptidases, a basic or neutral one,
and an acidic one inactive at pH 4,4 could be discerned, whereas in
microsomal fractions a broad specificity aminopeptidase preferring
methionine was detected.
5. There is a considerable degree of similarity in the pattern of
aminopeptidases within different blood cells. This may suggest that their
functions are correlated to the physiological role of a particular cell
compartment, rather than to that of a distinct cell type.

Summary: 1. After being treated with nonopsonized zymosan A or
phorbol-12-myristate-13-acetate, human polymorphonuclear leukocytes release
aminopeptidases together with granules marker-enzymes and vitamin
B12-binding protein.
2. Chemotactic peptide, fMet-Leu-Phe and its analogs, fMet-Phe,
fMet-Ala and fMet-Leu-Phe-Lys have a similar effect.
3. By their isoelectric point determinations the released
aminopeptidases correspond to the enzymes from granules. Among
aminopeptidases released the highest activities were those toward
methionine- and alanine-2-naphthylamide and the lowest one toward
arginine-2-naphthylamide.

Summary: A UV-inducible, damage-specific DNA-binding (DDB) protein
with high affinity for double-stranded UV-irradiated DNA has been
identified recently in monkey kidney (CV-1) cells. We have now purified the
DDB protein from extracts of CV-1 cells using hydroxylapatite,
phosphocellulose, Mono S, and DNA-affinity column chromatography. The DDB
activity, either from mock-treated or UV-induced cells, is heterodisperse
in column chromatography, and separation of three forms of the protein was
obtained on a phosphocellulose column. Analysis of purified preparations by
sodium dodecyl sulfate-polyacrylamide gel electrophoresis demonstrated that
>90% of all three forms is a protein of about 126 kDa. The size of the
native DDB protein was deduced from gel filtration and native
polyacrylamide gel electrophoresis to be 210 kDa, which suggests that the
native DDB protein in solution is a homodimer. Preparations of partially
purified DDB protein from UV-treated cells have enhanced levels of DDB
activity and the protein, when compared with similar preparations from
mock-treated cells. This damage-recognition protein, alone or in
conjunction with other subunits, may be of general importance for the
initial recognition of DNA damage in mammals.

Summary: Based on the kinetic constants determination and the
mathematical model of the reactor system developed, the performance of
axial flow packed bed continuous enzyme reactor system was studied
experimentally and also simulated with the aid of a computer for ultimate
objective of optimization of the glucose isomerase reactor system. A
reactor model was established analogous to heterogeneous catalytic reactor
model taking into account the effect of fluid mass transfer and reversible
kinetics. The investigated catalyst system consists of immobilized
Streptomyces bambergiensis cells containing the enzyme glucose isomerase,
which catalyzes the isomerization of glucose to fructose.

Summary: A purification procedure of an extracellular alpha-amylase
from Streptomyces rimosus, oxytetracycline-producing strain, is described.
The enzyme obtained was shown to be an acidic (pI 4.75) monomer with a
relative molecular mass (Mr) of 43000, containing three cysteines involved
in the catalytic activity of the enzyme. Its amino-terminal part has 57-67%
homology with amylases from other Streptomyces species. S. rimosus
alpha-amylase is sensitive to higher temperatures, and partially stabilized
by Ca2+ ions. It hydrolyses starch (optimum at pH 5.0-6.0) in an
endohydrolase manner giving rise to maltotriose, maltotetraose and higher
oligosaccharides. Starch granules, except those from rice, were not
significantly affected by the isolated alpha-amylase.
