Summary: Among many viral diseases, the acquired immunodeficiency
syndrom (AIDS) has evolved despite intensive research on prevention and
treatment into a major epidemic in the last decade. Due to intensive
efforts by chemists and molecular pharmacologists a number of nucleoside
analogues have been discovered to possess potent antiviral activities
against HIV the causative agent for AIDS. This investigation is conerned
with the synthesis of the novel nucleoside analogues containing exocyclic
pyrrolo moiety and acyclic side chains attached to the purine ring.
Conformation of the N-9 substituted regioisomers containing
2'-acetoxypropyl and 2'-propenyl chains was unambigously proved by their
X-ray structure analysis. The purpose of this work is to evaluate the novel
compounds on various viruses causing opportunistic diseases including HIV,
herpes and human hepatitis B virus.

Summary: In connection with our previous studies on
dibenzosemibullvalenes we have synthesized the novel regiospecifically
trifluoromethyl substituted dibenzobarrelene (2) and dibenzosemibullvalene
(3) for their evaluation on antidepresiveactivities. The stereostructure of
compounds (2) and (3) was deduced on the basis of their H-1, F-19 and C-13
1D and 2D NMR spectra. The magnitude and the multiplicity of C-F coupling
showea to be a good structural probe. The assignment of C-F multiplets was
corroborated by H-1 inversion (INCDQ) and NOESY spectra and by molecular
modeling. Three-bond C-F couplings range from 1.23 Hz to 5.6 Hz and they
are related to dihedral angles. Thelongest observed C-F coupling constant
was through five bonds, amounting to 1.48 Hz, at ortho carbons of
substituted phenyl ring in (3).

Summary: The synthesis of the novel chiral 2H-chromenes 7-9 is
described in this work. The purpose of this compounds preparation was to
separate their enantiomers and apply optically active thus obtained in
stereodynamic studies. Preparative resolution of enantiomers of 7-10 was
achieved by liquid chromatography on triacetylcellulose. The gibbs free
enthalpies of activation for interconversion of enantiomers in 7-9 were
determined by thermal racemiyation of preparatively separated enantiomers.
It was established that this reaction proceeds by the mechanism of
reversible electrocyclic opening and reclosure of the 2H-1-benzopyran ring.
It was confirmrd by X-ray structure analysis that di(2H-chromene-2-yl)
ether(10) exists in the meso form,i.e. (2s,2R')-10. Furthermore, the
analzsis of structural data showed that geometries of the two benyopzran
parts of which consists skeleton of 10 are not identical, what means that
this molecule is pseudosymmetrical. The meso-10 undergoes thermallz induced
ring opening and reclosure. By this reaction the meso-10 equilibrates with
racemic mixture of enantiomers (2S, 2'S)-10 and (2R, 2'R)-10 as confirmed
by 1H NMR spectroscopy and preparive separation of stereoisomers using
liquid chromatography on triacetylcellulose. In the part of studies
concerned with bridged biphenyls, a series of chiral 2,2'-singly and
2,2'-6,6'-doubly bridged biphenyls 18-21 were synthesiyed in order to
separate their enantiomers and determine the absolute configuration by
analysis of their circular dicroism (CD)spectra. Resolution of enatiomers
has been achieved for the thiepine sulfoxide 20, thiepine sulfone 21 and
dithiepine 18 by liquid chromatography on triacetylcellulose. The absolute
configuration of the bridged biphenyls 18, 20 and 21 was determined on the
basis of their CD spectra. Thus, it was established that the first eluated
(-)-enantiomer in 18, 20 and 21 correspond the S-configuration.
