    Summary: Isolated posterior gills of shore crab Carcinus maenas acclimated to brackish water were bathed with medium (200 mM Na) collecting the internal perfusate during subsequent periods of 5 min.During a single passage through the gill the pH of the perfusion medium decreased from 8.1 to 7.7, a result implying that the gill posseses structures of "recognizing" unphysiologically high pH values of the hemolymph and regulating them down to physiological values of 7.7. The calculated apparent proton fluxes from the epithelial cells into the hemolymph space amounted to 17,9 micromol/h per gram fresh weight, a value of only 3.8% of net sodium fluxes observed under comparable conditions.When 0.1mM KCN, an inhibitor of cytochrome oxydase, or 0.5 mM ouabain,a specific inhibitor of Na,K aTPase were applied in the internal perfusate down-regulation of pH was no longer observed and gill was completelly depolarized, i.e. transbranchial potential difference dropped from 7.8 to 0 mV. The results obtained suggest that pH regulation by crab gill depends on the intact metabolism of the branchial epithelium and that Na/H exchanger seems to be not involved in these processes.

    Summary: Spatial bacterial abundance and production in the stratified Krka River estuary were analyzed in the fall of 1990.The bacterial abundance within the estuary was several times higher in the open central Adriatic waters, but was lower than in the mostly eutrophicated western part of the northern Adriatic.Halved drop (35%) of bacterial number between the brackish and seawater layers compared to a 60 % decrease of the flagellates number indicated a net bacterial production below the halocline which grayzing was unable to match.A prevailing bacterial remineralization in the seawater layer is supposed.

    Summary: Sublethal and lethal toxicity tests of heavy metals on two species of marine invertebrates were performed.In addition, transport of Cs, Cd and Hg across the isolated gill epithelia,used as a modelling system in the metal transport was studied.Under the continuous flowing test conditions, lethal toxicity concentration of the methyl mercury (96h LC-50 = 0.031mg/l) is more than one order of magnitude lower than that for the mercuric chloride (96hLC-50=1.04 mg/l).Influxes of radioactive labeled mercurial compounds and caesium were studied across perfused isolated gills of the shore crab Carcinus mediterraneus.The results show an increased transfer of methylmercury over inorganic form across perfused epithelial cells.Caesium net transport was directed from the hemolymph side crossingm basolateral and apical surfaces into the bathing medium.Methylmercury compound had reduced transbranchial potential difference values (-3.6 mV in the control group) close to the zero. After 1-8 days of exposure of the sea urchins Sphaerechinus granularis in the 10 mg Cd/l sea water, gametes were isolated and fertilized. Most of them (3-8 days exposure) exhibited delay in development, irregular fertilization membrane formation in the early prism stage and swimming blastula.

    Summary: Results concerning determination of Cs-137 and other artificial and natural radionuclides in Northern and MiddleAdriatic samples of seawater (at different depths) plankton, pelagic,benthic organisms and sediments, obtained by means of gammaspectrometry, are presented and discussed in this paper. Different marine specimens have been collected since 1979 during Oceanographic field trips executed within the ASCOP(Adriatic Scientific Cooperative Program) with Italian and Croatian research ships. Right after the Chernobyl event (26.04.1986) significant sampling and radionuclides determination have been made on marine species (1986-1987). The results allow to underline a representative picture of theCs-137 concentrations in the considered sea water, plankton,organisms and sediment samples.

    Summary: The activity of enyzme carbonic anhydrase in mollusc Mytilus edulis from the early embryonic stage to the formation of veliger larvae was studied.The present results indicate that maximum carbonic anhydrase activity in the D-shaped veliger at the end of forming of prodissoconch is correlated with biomineralization process of bivalve shells.

    Summary: Crustacean gills are multifunctional organs operating in exchanges of respiratory gases, osmo- and ionoregulatory active uptake, regulation of the acid-base status of the hemolymph, and excretion of nitrogenous metabolic end products. Following previous work on active ion uptake across crab gill this work is devoted to pH -regulation by and excretion of ammonia across the gills of the shore crab Carcinus maenas.We were interested to answer whether H and/or HCO3 (pH regulation) and NH3 and NH4(N-excretion) are regulated across the gill epithelium using separate mechanisms or utilize existing pathways for ion translocation. Isolated posterior gills of brackish-water (10 x 10-3, salinity) shore crabs were perfused and bathed with dilute sea water (200mM Na). We observed that the pH of the perfusate (pH=8.1) after a single passage through the gills was regulated down by ca. 0.4 pH units exhibiting physiological means of pH 7.7. Regulation of pH was suppresed when 0.1 mM KCN or 5 mM ouabain, aspecific inhibitor of Na,K ATPaze, was applied in the perfusate.Regulation of pH was also suppressed when 10-4 M acetazolamide was applied in the perfusate.Regulation of pH remained unchanged when 10-4M amiloride or 10-5 M EIPA inhibitors of Na channels andNa/H exchangers, were applied in the bath and in the perfusate orin the bath alone.The results suggests that pH regulation depends of the Na,K ATPase in cooperation with a basolateral Na/HCO3 from hemolymph into the epithelial cell utilizing the existing Na gradient and thus decreases hemolymph pH. Findings in this work suggests that nitorgen is excreted in the form of NH4 ions partially via the basolateral Na,K ATPaze and as a major proportion via apical nonspecific Na channels.

    Summary: Electrical resistance of the gill lamella and the intracellularacidification of the Carcinus gill cells were studied. Theposterior gill lamella resistance measured by the Ussings type ofchamber was 60 Ohm cm2. The lamella resistance is significantlyincreased in the presence of 10-4M amiloride in the bathingsolution. The lamella was splitted out to make a feasible access to thebasolaterally oriented cell surfaces for the intracellularacidification measurements. For this purpose the cells wereloaded in the fluorescent dye BCECF and acidified by 30 mM NH4Clpulses.After a sequential cell washing by different solutions theintracellular acidification rate was measuredmicrospectrofluorimetrically.New informations about sodiumdependent and amiloride non-sensitive intracellular acidificationof the Carcinus gill epithelium cells were described. Additionally,in the isolated gill preparation perfused and bathedwith the identical dilute sea water medium,acidification of thecollected perfusate was observed. The acidification rate was notaffected by the EIPA (amiloride derivative), a strong inhibitorof Na/H exchanger.The cuticle and the underlying epithelium celllayer are two selective barriers for the osmolytes of thecrustacean gills. The results suggest the presence of Na/HCO3exchanger, a mechanism which is located probably on thebasolateral side of the Carcinus gill epithelium.
