Summary: Ubiquitination of proteins is a crutial step in the controlleddegradation process of many polypeptides. Here we show thatsponges, the simplest multicellular group of eukaryoticorganisms, are also equipped with the ubiquitin pathway. Thepolyubiquitin cDNA was isolated and characterized from the marinesponge Geodia cydonium. The open reading frame contains sixubiquitin moieties, which are lined up head to tail withoutspacers. A comparison of the predicted amino acid sequence of thesix sponge ubiquitin-coding units with those from other organismsrevealed a high degree of homology (>93%). The ubiquitin gene isexpressed to almost the same extent in the two main compartmentsof the sponge, the cortex and medulla. However, only in thecortex are detectable amounts of the ubiquitin proteinsynthesized. The ubiquitin protein isolated from the spongeorganism was found to initiate protein degradation in theheterologous reticulocyte system in the same manner as bovineubiquitin.
