    Summary: Two bifunctional plasmid vectors pZG5 (7.45 kb) and pZG6 (6.95kb), for gene transfer between Streptomyces spp. and Escherichiacoli have been constructed by fusion of the multicopybroad-host-range Streptomyces plasmid pIJ350 with E. coliplasmids Bluescribe M13- (pZG5) or pUC18 (pZG6). Both plasmidspossess several unique restriction sites suitable for DNAcloning. Stable transformants of Streptomyces rimosus R6 and S.lividans 66 were obtained, harboring intact plasmids regardlessof colony age or multiple subculturing. Moreover, pZG5 and pZG6were successfully used to introduce several homologous transferRNA genes into S. rimosus.

    Summary: We demonstrate that the cells of the sponge Geodia cydonium areequipped with the basic elements required for a retinoic acid(RA)-dependent response pathway; RA was identified andquantitated, the cellular RA-binding protein (CRABP) was detectedand the nuclear RA receptor (RAR) was found. In the isolated cellsystem the level of CRABP, but not of RAR, is strongly inducedafter incubating the cells for 10h with the homologousaggregation factor. In induced cells incubation with 0.3 microMRA results in a strong down-regulation of the c-myb (orc-myb-related) proto-oncogene (M(r) 63,000; mRNA 3.3 kb). Wepostulate that this pathway is also functionally active and thatRA acts as a natural morphogen.

    Summary: Streptomyces, Gram-positive mycelial soil bacteria with complexlife cycle and morphological differentiation, posses severalclasses of promoters which are recognized by different sigmafactors during the process of contolled gene expression.Regulatory sequences involved in gene expression in S. rimosusare not known and there are no reports about the mode ofexpression of tRNA genes in Streptomyces. We recently completedsequence analysis of rrnF operon from S. rimosus and found onlyone (P4) promoter in front of this operon. We also analysedexpression of seven tRNA genes from S. rimosus, two independenttRNA-fMet genes and a cluster of five Gln-Glu genes. All threetRNA gene promoters are quite diferent. Deletion experimentsshowed that tRNA genes in the clusted are all cotranscribed fromthe same promoter localized 140-65 bp upstream from the firstgene where the sequence TTGCGC-17-TAGACT was found. This tRNAgene promoter belongs to the class of Streptomyces - E. coli typepromoters (SEP).

    Summary: The marine sponge Geodia cydonium contains several lectins. Themain component, called lectin-1, is composed of three to fouridentical subunits. The subunits of the lectins were cloned froma cDNA library; two clones were obtained. From the deduced aasequence of one clone, LECT-1, a mol.wt of 15313 Da iscalculated; this value is in good agreement with massspectrometric analysis of 15453+-25 Da. The sequence of anotherclone, LECT-2, was analysed and the aa sequence was deduced(15433 Da). The two subunits have a framework sequence of 38conserved aa which are characteristic for thecarbohydrate-binding site of vertebrate S-type lectins.Clustering of lectin sequences of various species following theirpairwise comparison established a dendogram, which reveals thatthe sponge lectin could be considered as the ancestor forvertebrate S-type lectins.

    Summary: The interaction of pyronin Y, an RNA intercalating drug, with thebinding of Rev protein from human immunodeficiency virus type 1(HIV-1) to Rev responsive element (RRE)-containing env RNA wasstudied. In a gel retardation assay, recombinant Rev proteintightly bound to in vitro transcribed RRE RNA. At a concentrationof 1ug/ml, complex formation was almost completely inhibited.Electron microscopy revealed that Rev oligomerizes in thepresence of RRE-containing RNA with the formation of shortrod-like structures of long filaments, depending on the length ofthe transcript. The assembly of Rev protein along RRE-containingRNAs was abolished after addition of pyronin Y. Thus pyronin Yrepresents the first compound described to inhibit Rev-RREcomplex formation under in vitro conditions.

    Summary: DNA-binding assays using mobility shift polyacrylamide gelelectrophoresis revealed the presence of a protein thatspecifically bound to a restriction fragment -288 to -191 bpupstream from the transcriptional start point of strR, aregulatory gene for streptomycin biosynthesis in Streptomycesgriseus. The binding site corresponded to an upstream activationsequence predicted from the results of in vivo promoter assays.The binding was greatly enhanced by 5 mM Mg++. This binding wasdetected with the protein source only from the wild-type strainand not from an A-factor-deficient mutant strain. The exogenoussupplementation of A-factor to the A-factor-deficient mutantstrain caused the appearance of the protein in the protein in theDNA-binding assay. A synthetic nucleotide 52 bp in length (regionfrom -293 to -242), which was synthesized on the basis of dataobtained from both retardation assays with dissected DNAfragments and in vivo promoter assays, was retarded by theA-factor-dependent protein. In addition to thisA-factor-dependent protein, at least three proteins withdifferent recognition site affinities capable of binding to theupstream region of the strR promoter were detected. The regionbound by these proteins showed anomalous electrophoreticmobility, like that of a bent DNA molecule. This region was foundto be transcribed in the orientation opposite to that of strR.

    Summary: One Streptomyces rimosus ribosomal RNA gene set (rrnF) was clonedinto pBR322 as 8.8 kilobase-pairs BamHI fragment and sequence ofthe 3' end of the operon with 5S rRNA gene was determined. 5SrRNA gene is 120 bp long and highly homologous with two otherknown 5S rRNA from Streptomyces. Remarkable sequence homologywith S. ambofaciens rrnD operon exists in the 3' noncodingregions, including first transcription termination signal. S.rimosus rrnF operon contains second putative terminator which isabsent in S. ambofaciens rrnD operon. tRNA genes were not foundat the 3' end of rrnF operon. The number of rRNA operons in S.rimosus was defined by Southern hybridization analysis. S.rimosus possess six rRNA operons and all rRNA operons containidentical PstI fragment of 4.5 kb. rRNA genes in operons areseparated by short intergenic regions and organized in the order16S-23S-5S. Efforts to connect tRNA genes with S. rimosus rRNAoperons were unsuccesful.

    Summary: In this work, expression of S. rimosus tRNA genes has beenstudied in vivo in homologous system (S. rimosus) and inheterologous system (E. coli). Seven tRNA genes were analyzed:two independent genes for tRNA-fMet and a cluster containing two tRNA-Gln and three tRNA-Glu genes. All S. rimosustRNA genes were (re)cloned in different combinations andorientations, with or without potential promoter regions, intostable bifunctional multicopy plasmid vectors pZG5 and pZG6,especially constructed for this purpose. Recombinant bifunctionalplasmids with tRNA genes were introduced into both hosts. Totalcellular tRNAs were isolated from S. rimosus and E. colitransformants containing different recombinant plasmids andsubjected to northern hybridization analysis for the estimationof the relative amount of tRNA-fMet, tRNA-Gln and tRNA-Glu indifferent cell lines. Northern hybridization analysis shows thatall analyzed promoters are active in homologous system. Fromthe deletion assay it can be concluded that all tRNA-Gln andtRNA-Glu genes are cotranscribed from the same promoter located140-65 bp upstream from the first gene. Sequence of this promoteris TTGGAC-17-TAATGT. In the heterologous system (E. coli) onlythe cluster of Gln-Glu tRNA genes is properly expressed.

    Title: Structure of rrnF ribosomal RNA operon from Streptomyces rimosus

    Summary: The primary structure of the S. rimosus rrnF ribosomal RNA operonhas been determined. This rRNA operon is located on the 8.8 kbBamH1 fragment. Deduced nucleotide sequence ( 6067 bp)contains a promoter region, the genes for 16S, 23S and 5Sribosomal RNAs, intergenic regions and termination sequences.This results represents the first primary structure of one S.rimosus RNA operon, and one of only few structures of rRNAoperons in Streptomyces. The 16S rRNA gene is 1529 bp long and highly conserved inrelation to four known 16S rRNA genes from Streptomyces. Thisgene contains highly variable regions which are specific forS. rimosus. Region between 16S and 23S rRNA genesis 298 bp long. The 23S rRNA gene is the longestsequenced gene in prokaryotes (3121 bp), as a result of theinsertions in the 5-end of the 23S rRNA gene. The region upstreamfrom the 16S rRNA gene, 689 bp long, contains potential 3- end ofone open reading frame ( last 61 amino acids). The potentialpromoters were found in the region 268-297 bp (P4) and420-448 bp (P3?) upstream from the 16S rRNA gene. Only P4promoter shares similarity with other known promoters of rRNAoperons in Streptomyces. Region downstream from P4 promoter iscomplementary to the intergenic region between 16S and 23S rRNAgenes and most probably play an important role in theantitermination of the transcription and processing of theprimary transcript. In this region are found highly conservedsequences known as A blocks.
