Summary: In control condition of in vitro culture the
differentiation and regeneration of plant organisms will be investigated
(Cucurbita pepo, horse radish and others). Gene activity and nuclear
genome in determined phases of growth and development will be observed.
Biochemical and gene manipulation with using Ti and Ri plasmids will be
tested.
Keywords: Regeneration, somatic embryogenesis, transformation, regulation of gene expression during regeneration and transformation, morphogenesis, differentiation, dedifferentiation, organogenesis, habituated tissues and cells, transgenic plants, modification of secondary metabolite production in cell cultures.
Research goals: Goal Establishment the role of endogenic and
exogenic factors (plant growth regulators, physical conditions, genom
modification) in physiological and genetical regulation of organogenesis
and somatic embryogenesis in plant cell and tissue culture in vitro.
Result - Conditions for the establishment, multiplication and adventitions
root induction of juvenile Quercus robur plants; for establishement of
whild cherry primary culture, multiple shoot induction and plant
regeneration in Betula pendula, the effect of genotype on Gerbera shoot
multiplication, influence of meristem culture and virus elimination on
phenotypical modifications of grapevine (V. vinifera L. cv. Refošk) were
found. - Peroxidases were studied in plant species in relation to
differentiation process and tumor transformation. Distinctive markers of
morphogenesis and tumori genesis were found. In habituated pumpkin callus
lines an increase in peroxidase activity meant that the embryogenesis was
triggered. Crown-gall and hairy roots were produced in horse-radish and
pumpkin explants. Parameters for pumpkin embryo maturation and unlimited
production of normal plants from very long-term embryogenic cultures were
analysed.
