    Summary: Skeletal muscles are the generators of active force and therefore the actuators in the locomotor system. Muscle tissue, namely, is characterized by properties like excitability, conductivity, elasticity and above all contractility, enabling the muscle the role of actuator. When researching muscle and muscular system it is important to explore the relation between the muscle's structure and function. We have pursued a study along these lines by providing morphohistochemical analysis of human muscle biceps brachii and electromyography (EMG) and dynamometry measurements of actively contracting muscle. Plotting the histograms of occurence of fiber diameters, at the scale of 3 micro meters (um) resolution, reveals that in younger persons the biceps brachii specimen for ATP-ase activity at pH 4.3 show a greater frequency of type II fibers with a diameter from 91 to 106 um, while type I fibers have the highest frequency of occurence at diameters ranging from 22 to 28 um. In older individuals the type II fibers are smaller with the highest frequency of fibers with diameters ranging from 61 to 79 um; the type I fibers are larger, with the highest frequency of fibers with diameters ranging from 58 to 76 um. Despite significantly different levels of maximum isometric force developed by our subjects - corresponding to their physical abilities - in all cases analyzed a proportionality between measured force and smoothed EMG values may be observed. The current level of scientific knowledge suggests with high probability, the existence, in subjects manifesting higher isometric force gradients, of a higher proportion of type II fibers characterized by higher contraction strength.

    Summary: The aim of this study was to determine whether injuries of the N. ischiadicus of IV and V degrees (by Sunderland) provoke differences in the growth rate of nervous fibers, and changes in types of muscular fibers in 2-month old AO rats. The IV degree injuri was done by nerve compression with microneedleholder for one minute. While the V degree injuri was done by transection of the N. ischiadicus in the other group. By observing the muscle weight, it was established that in the first group of animals subjected to compression of N. ischiadicus, the muscle weight decreased until the 17th day when there was no further decrease of muscle weight . Muscle weight continually decreased until the end of the experiment in the second group of animals with V degree injuri. Histological examination of nerves, established that the distal axons in both cases were affected by Wallerian degeneration. The IV degree injuri left the epineurium intact, unlike the V degree injuri which damaged it. The initial signs of regeneration of the nervous fibers were presented in both ways of denervation, compression and transection alike. Muscle fibers of M. soleus and M. tibialis anterior were typified using the method for demonstration of myosin ATP-ase activity. In M. soleus, characterized by prevalence of type I fibers, the number of type II fibers gradually increased. Regarding ATP-ase activity, we established that changed muscle fiber types in soleus muscle were presented in both types of denervation, indicating type I fibers to be under greater neural control than type II muscle fibers.

    Summary: By chronic electrical stimulation elicited changes are different, depending on frequencies, regularity and duration of the stimulation. Mainly, the results show a fast to slow fiber transformation. We were interested in how pathological conditions of repeated tonic-clonal covulsions influence to morphohistochemical characteristics of the deep dorsal muscle (DD) and diaphragm. Type I and II fibers were examined by myosin ATP-ase method. The connective tissue of the endomysium and perimysium was stained by the Weigert-elastica-Van gieson method. Female rats were stimulated with electrodes attached to the ears for 5, 10 or 15 days. The applied stimulus was 75 mA, 50 Hz, lasting for 1 second. Such stimulation produced tonic-clonal convulsions lasting 12-15 seconds and opistotonus for 7-10 seconds. Type II fiber predominated in medial and lateral part of the DD, and in the lateral part type I fiber is less than in the medial part. A 5- or 10-day stimulation does not induce changes in distribution of fiber types in the DD and diaphragm, but induces a decrease of oxidative capacity of type I fiber and their number. A 15-day stimulation induces a decreased of the number of type I fibers in most parts of the cross section area but at some points can be seen transformation type II to type I fibers. The endomysium is visible and thick.

    Summary: Oxidative and glycolytic capacity of muscle can indicate its functional properties and can also be presented by enzyme-histochemical methods. This analysis was carried out on samples of 36 human multifidus muscles taken at obduction from the level at L4, in persons who died of sudden death aged from 18-50 years. The type I fibers, making up 61.52% of all the fibers, show stronger oxidative capacity, while the activity of NADH-TR in the type II fibers is weaker. The type II fibers demonstrate a stronger glycolytic capacity than the type I fibers. Only a part of type I fibers in some samples shows a somewhat stronger activity of alfa-GPDH, but weaker than that of the type II fibers. In a few samples one part of the type I fibers shows a somewhat weaker NADH-TR activity than the other type I fibers in the same sample, but is stronger than that of type II fibers. There are no type II fibers with a stronger oxidative capacity than that of the type I fibers.

    Summary: The aim of this study was to investigate the metabolic activity of axial sceletal muscle fibres in two types of telost from the Adriatic Sea - the catfish (Scyllum canicula cuv.) and the sardelle (Clupea sprattus L.) using the following histochemical techniques. The NADH-TR method by Spanhoff is used to prove enzymatic activity of the respiratory chain. The alfa-GPDH method by Weiner shows the intensity of enzyme activity of glycolysis. In applying the PAS method for carbohydrates, and Sudan black B for lipids we can also notice differences which depend on the type of muscle fibres. Our results show a high activity of the Krebs' cycle enzyme (NADH-TR) in type I fibers. This type are fibres of small diameter with a well developed endomysium, localized laterally, below the skin in both species. The same applies to results obtained by the PAS and Sudan black B methods, which indicate higher levels of carbohydrates and lipids in type I fibers in sardelle, as compared to those in the catfish. Type II fibers form the main body mass in both species. These fibers have wite diameter, with a poorly developed endomysium around them. They are NADH-TR negative and alfa-GPDH positive.

    Summary: The fiber diameter, as one of the morphometric parameters, is mentioned in respect to fiber types. Thus, the diameter of type I fibers is smaller and of type II is greater, although it is evident that the diameters of both muscles fibers differ considerably within a group. Therefore, the study was focused on the distribution of all muscle fibers in respect of their diameter, separately for both types. On muscle samples of the biceps brachii obtained from six healthy persons aged 20-40 yr, quantitative and histochemical analysis was performed. Considering the myosin ATP-ase activity (pH 4.3) fibers were divided into dark (type I) and light (type II). Their number was determined and the diameters measured. The distribution frequency of all dark and light fibers diameter with a step of 3 um shows that there are 41 morphofunctional units (MFUs) in the biceps brachii, indicating their great heterogeneity. The presence of dark fibers of small diameter (22-31 um) is marked (28%), cleary indicating their frequent activity at arm movement with small muscle force, while fibers of greater diameter, being dark or light, are successively activited when greater or great muscle force is required. The distribution frequency of both fiber types in respect of their diameter, demonstrates marked heterogeneity of fibers and plasticity of there function.

    Summary: Male rats, y-line were treated 10 days by time-varying, pulse, low frequency, magnetic field. We wanted to fortify the influence of magnetic field density 10 mT, with frequency of 10 Hz, 10 minutes daily, through 10 days continuously, on denervated skeletal muscles. Denervation was performed by compression of sciatic nerve and after that rats were stimulated. On transverse sections of soleus and tibialis anterior muscles we wre represented the types of muscle fibre and capillaries by histochemical methods. The biggest change in stimulated muscles was increasing the number of capillaries. That number is greater in healthy or denervated muscles which were stimulated, than in healthy or denervated controls. Measuring the muscle mass we noticed decreasing the percentage of atrophy in stimulated muscles after 18 days of denervation.

    Summary: By electrical stimulation (75mA, 50Hz, lasting 1sec) wereproduced tonic-clonal convulsions with expressed opistotonus.Such stimulation was repeated daily in the course of 5, 10 or 15days. Serial sections were stained for myosin ATP-ase at ph 9.4and 4.3, with NADH-TR and alfaGPDH. Type II fibers predominate indeep dorsal muscle (DD) while fibers type I are rare and single.But most of the fibers are positiv with NADH-TR method what isnot in correlation with results of mATP-ase activity. Conversion and clustering of fibers type I isseen after 15 days at some places in DD muscle. Endomysium andperimysium are more thicker.

    Summary: Muscle samples of m.multifidus end m.longissimus were taken inthe level of 4th lumbar vertebra from persons aged from 18 till60 years. On transverse serial sections distribution of musclefiber types was reaalized by method of myosin ATP-ase at pH 9.4.In m.longissimus percentage of fibers type I is greater than inm.multifidus but both muscles contain greater number of fiberstype I than type II.     Summary: We investigated the development of muscle fiber types in fetaland newborn m.erector sspinae by method for myosin ATP-ase. Theresults show that all fibers are undifferentiated IIC fibersuntil 20. week of gestation. Until 30. week appear big, singlefibers type I surrounding with little undifferentiated fibers.After 30. week the number of fibers type I increases, otherfibers are differentiated as type II and a small number of stillundifferentiated fibers are presented. During first year of lifedifferentiation of fibers type II is going on.

    Summary: Paravertebral muscles are one of the very important factors in maintaining the vertebral function. In the patients with low back pain their role is even more important because serve as compensatory mechanism. We analysed the density and the area of cross sections of paravertebral muscles at the level of the intervertebral spaces L4/L5 and L5/S1. We correlated our results with age and pathological changes of the examined vertebral segment.In analysis of the involutiv muscle changes degree, we concluded how is very important to include the fisical activity of the patient before the illness and for how long time the symptoms are presented. So the results showed that muscle changes are more dependent of muscle fisical activity before the illnes and the lasting of symptoms which couse the changing vertebral motility, than of the pathological changes of the vertebral segment.

    Summary: Controversy surrounds the use of optic magnification for peripheral nerve surgery. The aim of this study was to distinguish the eventual differences in regeneration, measured as motor nerve conduction velocity (MNCV), compound muscle action potential (CMAP) amplitude and the clinical test equivalent (CTE), of loupe magnification vs. microscope assistance tibial nerve grafts. Four groups of 12 animals in each, were operated upon with 2.5 x, 5 x (loupe), 16 x and 40 x (microscop) magnification. The recovery results were assessed by 2 independent evaluators 5 months later. MNCV measured on the operated nerve was expressed as a percentage of the normal side, while CMAP wave lenght, latency period, amplitude and results of CTE were presented as arithmetic mean. In each animal CMAP wave was biphasic, spread over 2.42+_1.08 msec, with latency period of l.78+_0.70 msec. MNCV in the group operated with loupe magnification and microscope assistance was 26.77+_9.37 m/sec and 44.19+_11.36 m/sec respectively. The same scores expressed as a percentage of the normal side are 39.37% and 63.90% (p<0.05). CMAP amplitude was 5.53+_2.78 mV in the loupe group and 7.01+_2.24 mV in microscope group (p<0.05). CTE was 2.0/3 in loupe and 2.8/3 in microscope group (p<0.01). Our results suggest the following: - regeneration plateau has been reached as indicated by CMAP wave pattern, length and distal motor latency period - the fiber diameter is higher in the microscope group as indicated by difference in MNCV - the CTE difference confirms better motor-unit reinnervation coordinated by cortically integrated sensory feedback in the microscope group again.

    Summary: Due to various descriptions of deep dorsal muscles in literature,gross dissection was done on embalmed cadavers in order toestablish relations between these muscles as well as all otheranatomical characteristics. The lumbar parts of m.multifidus and m.longissimus thoracistaken from the level of the fourth lumbar vertebra were alsohistochemicaly analysed. The results show a predominance of type I fibers in both muscles.The diameter of these fibers is significantly greater than thatof type II fibers. The glycolytic capacity is strong in type IIfibers while oxydative capacity is strong in type I fibers.The predominance of type I fibers, strong oxydative capacity inthose fibres and their greater diameter as compared to type IIfibers, point to the permanent role of those both muscles inmaintaining the lumbar lordosis, i.e. their primarily tonicactivity.

    Summary: The differentiation of fibre types in m. erector spinae of fetusand newborns was studied. The enzymes activity of mATP-ase showedonly the undifferentiated type IIC fibers before the 20th week ofgestation. Before the 30th week, scattered large type I fiberswere differentiated. After the 30th week fibres type II werepresented. At birth, we found a similar percentage of fibres typeI and type II with still a significant great number ofundifferentiated type IIC fibres. During the first year of lifedifferentiation of fibers type II was going on. The activity of oxidative and glycolytic enzymes was very weakduring fetal development.     Summary: Skeletal muscle can be transfected in vivo with plasmid desoxyribonucleic acid, but the efficiency of gene transfer is relatively low. We have compared the efficiency of direct gene transfer in normal and regenerating rat skeletal muscle. The muscle necrosis and regeneration was induced by an intramuscular injection of bupivacaine in the soleus muscle of adult rats. Plasmids containing beta-galactosidase or chloramphenicol acetyl transferase genes, driven by viral promoters were injected three days after bupivacaine treatment into the regenerating and the contralateral uninjured muscles. The expression of cloramphenicol acetyl transferase activity was 87-fold higher in regenerating, compared to control muscles 7 days after the transfection, but decreased at 30th and 60th day. The Southern blot analysis showed that the predominant form of cloramphenicol acetyl transferase desoxyribonucleic acid in transfected muscles was episomal; however, cloramphenicol acetyl transferase activity measurements, performed on the same transfected muscles, showed no correlation between the enzymatic activity and the amount of plasmid desoxyribonucleic acid. The expression of beta-galactosidase was detected in numerous regenerating fibres of the injured soleus muscles at the 7th day post-transfection. In contrast, only few positive fibers were found in control muscles. Focal infiltrates of the mononuclear cells, surrounding and invading selectively beta-galactosidase positive fibers, were observed at 30th day post-transfecton. This may suggest that an immune mechanism is implicated in the progressive loss of transgenes over time. The finding that regenerating muscle fibers display a higher efficiency of transfection may be relevant to gene therapy of Duchenne muscular dystrophy, since regenerating fibers are numerous in the early stages of the disease.
