Title: Production of leukemia inhibitory factor mRNA and protein

Summary: Both human and rodent immortalized and malignant bone
cellsexpressed a single 4.4 kb mRNA transcript that hybridized to ahuman
LIF cDNA probe in Northern blots. LIF mRNA was undetectablein unstimulated
rodent osteoblast-like cell lines, but treatmentwith LPS, TNFalpha, TGFbeta
or inhibitors of protein synthesisinduced the expression of LIF message.
Primary osteoblastenriched cells did not express LIF mRNA aither
constitutively orafter TNFalpha or cycloheximide. LIF protein was detected
byradioimmunoassay constitutively in the conditioned medium fromhuman
osteosarcoma cells, and TNFalpha or PMA stimulated therelease of LIF
protein. These results show that different humanmalignant and rodent
immortalized clonal bone cell lines canexpress and regulate steady-state
LIF mRNA and produce LIFprotein but primary cultures od fetal at
osteoblastic cells donot express this cytokine. Hence, LIF may regulate
malignantosteogenic cell growth and function but not normal bonemetabolism.

Summary: The development of acute phase response (APR) and bone loss
inrats with talc-granulomatosis was studied in vitamin D-deprivedrats.
Serum APR in rachitic rats consisted of hypozincemia,hypercupremia,
increased alk. phos. activity and ACTHconcentration, similar to control
animals with inflammation.Rachitic rats had decreased peripheral blood cell
counts but notthe splenomegaly of control animals with inflammation.
Incontrast to the controls, rachitic rats did not show loss oftrabacular
bone or decrease in osteoblast number in tibialmetaphysis, but only
abundant osteoid on bone surfaces. Theseresults indicate that vitamin D is
necessary for the developmentof the APR during inflammation, particularly
in the induction ofspleen and bone changes. The discrepancy of blood on one
and boneand spleen indices of APR on the other hand indicate that theremay
be divergent pathways in the generation of inflammatoryresponse, some of
which may be dependent on vitamin D.
