    Summary: This university textbook primarily has been intended for use inpractical teaching of veterinary microbiology and immunology.     Summary: The main traits of coagulase-positive and coagulase-negative staphylococci findings in inflammed cow udder are presented. The paper also presents the results of sensitivity tests of different strains of S. aureus to some penicillin antibiotics, i.e. penicillin G, ampicillin and cloxacillin. Determined MICs of individual antibiotics showed that all tested strains were susceptible to cloxacillin. Only 8 strains (8.9%) showed resistance to penicillin G and ampicillin. MICs of cloxacillin ranged from 0.062 to 1.0 mcg/ml. Resistance to penicillin G and ampicillin was result of formation of bacterial penicillinase which was found in all resistant strains.

    Summary: Microsporum canis infection in a male young lion (5-month old) originating from the Zagreb Zoo was diagnosed. The lion was kept in a household as a pet. Circular and oval scaly skin lesions with alopecia (mostly on lion's legs) were observed, but the origin of the infection was not determined. The lion's temporary owner suffered from the same ringworm, probably transmitted from the infected young animal.

    Summary: Exudative epidermitis (EE) in piglets occurs in peracute, acute and subacute form and it is sometimes characteriyed by high morbidity and sometimes high mortality. The most expressed changes in affected animals can be seen on the skin manifested by dermatitis of varing intensity and extend, accompanied severe exudation and exfoliation of epidermis and changes in the kidneys and ureters. The signs of disease are specially pronounced in piglets up to 10 days of age in wich a fatal ending is frequently recorded in spite of undertaken treatment. As the disease is highly contagious, it is indispensable to separate healthy from ill or suspect animals and to move them into other clean, dry and airy premises.

    Summary: Fourteen sera from dogs naturally or experimentally infected withMicrosporum canis were examined by ELISA in order to study thehumoral response to infection. A group of 12 animals (7laboratory dogs and 5 dogs from natural environment) was used ascontrols. ELISA OD values in twofold serial dilutions from 1:100to 1:51200 were compared between the groups. Significant antibodyresponese to infection with M. canis was observed in naturallyand experimentally infected animals and confirmed by tresholddetermination. Specificity for the dermathophyte fungi was testedby performing cross-reactivity assays with three fungal antigens,Malessezia pachydermatis, Candida albicans and Aspergillusfumigatus. There were no statistically significant indications ofcross-reaction, except with anti-A. fumigatus IgG, wheredifferences were noticed within the groups of naturally infectedanimals and controls. The use of ELISA technique insero-diagnosis of dermatophytosis in dogs is discussed.

    Summary: The occurrence of Mycoplasma pulmonis in five separate conventional breeding of laboratory mice and rats has been investigated. This mycoplasma was isolated from 15 (38.4%) of 39 examined mices and 43 (75.4%) of 57 examined rats. In infected facilities 66.6-90% mices and 62.5-100% rats were harbouring mycoplasma. Only in one stock acute form of infection was established. The results of this investigation clearly suggest that infection by M. pulmonis is very distributed in our breeding stocks of laboratory animals, but rarely manifested by apparently visible clinical and patoanatomical alterations. This may be the cause of mis judgement in explanation of results achived in biological trials.

    Summary: The efficacy of an inactivated and an attenuated vaccine against Aujeszky's disease (AD) has been evaluated in fattening pigs. Inactivated vaccine was prepared from strain B-KAL 68 and attenuated vaccine from Bartha strain. Vaccinated and control pigs were challenged intranasally with NIA 3 strain of AD virus. According to therma curves, weight losses and the time necessary for recovering the weight of pigs, the protection was good but not absolute in vaccinated pigs. Control pigs showed severe signsof AD affecting the respiratory and nervous system. Greater weight losses were found in pigs vaccinated with inactivated vaccine than in those vaccinated with attenuated vaccine.

    Summary: The first outbreak of rabbit haemorhagic disease in Croatia were detected at the end of 1989. In 1990, the disease was registered in 24 rabbitries in the south-western part of Croatia. At the same time the European brown hare syndrome was also frequently detected in free living hares (Lepus capensis) from Croatia. Both syndromes are documented on the basis of epizootiological, pathological, virological, and experimental transmission findings. Haemagglutination activity is detected in liver suspensions of naturally and experimentally infected animals. Calicivirus-like particles were detected as the caustaive agent.

    Summary: The paper describes the occurrence of Aujeszky's disease in hounds in Croatia. In total 9 dogs, eight duchs dogs and one pointer, have developed the clinical signs of Aujeszky's diseaseand 4 dogs died from peracute form. All dogs were fed with crude swine viscera. Diagnosis was confirmed by the isolation of herpesvirus on BHK-21 tissue culture. Pathogenicity of isolated virus was confirmed by test in rabbits.

    Summary: The sensitivity to a combination of trimethoprim and sulphonamide (TMP+SF) of 5,917 bacterial strains isolated from different animals over the period 1980-1992 and of 24,448 human isolates grom patients over the period 1983-1992 was tested by the agar diffusion method. The highest incidence of resistance to TMP+SF was recorded among animal isolates of E. coli (57,6%), Staphylococci (53,4%), especially S. intermedius strains (81,9%), and Streptococci (40,9%). However, over the period under observation, a significant increase in resistance was noted in all bacteria of animal origin that were examined. In general, strains of human origin were less frequently resistant to TMP+SF but increasing resistance was also evident among these isolates, with the exception of Campylobacter. The average percentage resistance for human isolates ranged from 2.5% for Salmonellae to 10.7% for Y. enterocolitica, 17.4% for E. coli, 17.8% for Shigellae and 23,3% for Campylobacters.

    Summary: In the paper literature data on the mechanism of the effects of trimethoprim and sulphonamides to bacteria are given. Indications and advantages of using the combination of these chemotherapeutics in veterinary and human medicine praxis are described. Special attention has been drawn to the reports got more often from all the world on resistancy spreading among bacteria to trimethoprim and sulphomamides, supported also by investigations carried out in our country. Although combination of trimethoprim and sulphonamides can still generally be encounterd in the efficient antimicrobial specimens suitable for peroral use, these chemoterapeutics in our country, especially in veterinary medicine praxis, should no longer be used without control.

Although kids and young goats are predominantly affected, sheep and cattle can be spontaneously, and swine experimentally infected. On the occasion of the first etiological diagnosis of that mycoplasmosis in Croatia, review on the literature data has been offered in this paper.

    Summary: Serological and biochemical properties were studied with 33 S. suis strains isolated from pigs in Croatia. Isolates were examined for the presence of group D antigens, using a commercial reagent for agglutination and all S. suis strains cross-reacted with group D antiserum. Of the 33 isolates 30 (90.9%) belonged to capsular type 2, two strains (6.1%) to capsular type 1/2 and one strain (3.0%) was untypable. Biochemical characteristics of all the strains were also investigated and they were found to be very homogenous and typical for S. suis. A CAMP-like phenomenon was observed in all capsular type 2 and 1/2 strains. This is the first report on isolation of S. suis from pigs in Croatia.

    Summary: Polymerase chain reaction (PCR) is the method for in vitro nucleic acid amplification. It is used as a means to multiply the copy number of the characteristic genetic elements of pathogenic agents. Since PCR is very sensitive and specific, it is frequently used to study pathogenesis and diagnosis problems of infectious diseases. It is suitable for the detection of pathogens that are difficult to isolate or identify with conventional techniques of cultivation, such as the causing agents of latent and slow infections or mycobacteria and chlamydia. General principles of PCR are given in that article.

    Summary: In order to provide an objective method for monitoring the kinetics of germ tube and hyphae formation under different conditions microscopic measurements to analyze separately each germ tube and hyphae on macroconidiae of Microsporum gypseum was utilized. The influence of the liquid culture medium , the temperature and aeration on germination and growth of five the highest pecentage germination was obtained in Yeme broth at 32 oC. The effect of temperature on growth was more pronounced than that of the media and growth in Sabouraud broth at 32 oC was always greater than in Yeme at 27 oC. The presence or absence of ribs in culture flasks had no significant effecton the growth of germ tubes and hyphae. Results of this study imply that analysis of germ tube and hyphae growth pattern on the basis of a variety of parameters is a useful research tool for determination of germination kinetics.

    Summary: Based on the literature data the incidence of Streptococcus suisin humans and animals is briefly reviewed. Serological and biochemical properties are also described as well as the procedures for the isolation, identification and serotyping of Streptococcus suis strains. Our own experiences of identification and serotyping of the first Streptococus suis strains isolated from pigs in Croatia are also presented.

    Summary: Several introductory facts about fundamnetal properties which an ideal contemporary veterinary vaccines should posses, are present. Basic presentation of the existing vaccines and some examples of their inovations, production and administration are given. Special attention is paid to deletion mutants which are used as marker vaccines against herpes virus infection and aromati deletion mutants against salmonellosis. Further, genetically engineered multicomponent virus-like particles and vector-based vaccines, i.e. live vaccines which contain one or more genes from heterologous microorganisms, are elaborated.

    Summary: This presentaion deals with the innovations in immunoprophylaxis of dermatophytes in animals, referring first of all to trichophytosis in cattle, horses and fur-bearing animals in which the diseases is caused by Trichophyton verrucosum, T, equinum and T. mentagrophytes. An attention is paid to the immune mechanisms which are the basis of immunoprophylaxis, and the results obtained on antigenic properties of some dermatophytes, especially of M. canis are presented. These investigations are essential for the successful immunoprophylaxis, because they enable the selection of the representative strains with suitable immunogenic properties.

    Summary: A total of 40 horses were vaccinated against equine influenza either with whole-virus vaccine (WVV) or with split-virus vaccine (SVV). HI antibody response was higher in horses vaccinated with SVV than in horses vaccinated with WVV. Significantly higher antibody response was induced by primary course vaccination with SVV than with WVV for subtypes possesing H3N8. Four weeks after primary dose and two weeks after a second dose the HI antibody response to WVV were low or undectable for H3N8 subtypes.

    Summary: In a large breeding colony of laboratory Wistar rats different materials from 97 female rats were cultured on mycoplasma media. The most animals were examined at the age of 9 months. Mycoplasma were isolated from a total of 47 (48.5%) of examined animals. All isolates were identified biochemically and serologically as Mycoplasma pulmonis. Except in lung samples and ear contents, this organism was found in 44.0% of examined uterine washings. The large percentage (48.2%) of mycoplasma infected animals harboured mycoplasma in respiratory and genital tract. Uterine colonization with mycoplasma was first recorded in a unmated three months old female. After mating the number of animals with mycoplasma in genital organs rapidly increased. Although the most animals were clinically healthy on the autopsies certain percentage of them exhibited lung lesions varying in severity and distribution. Most frequently the lung were bilaterally involved by mean of apical lobes atelectasis and sometimes macroscopically observable advanced bronchopneumonia. Patohistologically the lesions were characterized by peribronchial lymphocytic infiltration, bronchiectasis, and in some occasions suppurative bronchopneumonia. In mild cases only accentuated lymphocytic peribronchial and perivascular infiltration was present, with sometimes observable lymphoid follicles close to larger bronchi. The genital organs of infected females did not reveal pathological changes.

    Summary: The infections caused by Clostridium perfringens type C, the causative agent of necrotic enteritis (NE), are spread worldwide and in the intensive pig production they lead to considerable profit losses. The etiological dyagnosis of NE was set up on one large pig-breeding farm. To investigate the possibility of its immunoprohylaxis in the pigs, a dozen of pregnant Swedish Landrace gilts on that farm were given twice Dizevak-Pliva modified vaccine which contained inactivated cultures and detoxicated toxins (beta-and epsilon) of C. perfringens type C and D. The vaccine was primarily indicated for the prophylaxis of enterotoxaemia in cattle, sheep and goats. Antibodies for beta-toxin of C. perfringens type C at the farrowing were found in the serum samples from the vaccinated sows ranged from 9 to 26 IU/ml and in their colostrum were varying from 12.5 to 27 IU/ml. In some litters the average antibody titer in the serum of piglets aged 46 hours was 5.5. to 24 IU/ml, and in 7 days old animals 2.25 to 15 IU/ml. Antobodies were not found in the serum samples of the trial and control gilts taken before vaccination. Ten of 118 piglets of the vaccinated sows died during the first week of life. Microbiological and pathoanatimical analyses revealed that none of them had been affected by NE. During the same period out of 113 piglets of the unvaccinated sows 18 died, and NE was diagnosed in 11 of them. Carried out research showed that the farm was affected with NE, and that Dizevak-Pliva commercial bacterin-toxoid of toxigenic strains C. perfringens type C vaccine given to the sows established passive immunity against the infection with causative agent of NE.

    Summary: A collection of Malassezia pachydermatis strains, 100 isolated from dogs at 2 geographically separate locations, one from a cat, and 3 from young bears, have been karyotyped. Cellular nuceic acid was extracted from a selection of these srains, assayed for the presence of extra-chromosomal nucleic acid, studied by restriction fragment length polymorphism (RFLP) analysis and Southern blotting against a poly (GT) probe. All but five of the strains five of the strains shared a common six band karyotype pattern with the bands astimated to be , 800 kbp, 1250 kbp, 145o kbp, 1500 kbp, 1850 kbp, and 1900 kbp. In the remaining five strains the smallest 800 kbp band was replaced with a double band. Of the isolates tested 45 % were found to carry a 6.4 kbp RNA "plasmid" and 21% of the isolates tested also carried a 1.8 kbp RNA "plasmid" None of the isolates examined carried the 1.8 kbp RNA "plasmid" alone. The RFLP patterns produced by the restriction enyzme Hpall consisted of a limited number of bands which were suitable for visual comparison although 39 % of the strains analysed produced indistinguishable patterns. Southernblots of BgIII digests hybridised against a poly (GT) probe showed considerable variation and appeared to provide an ideal method of discriminating between the M.pachydermatis strains.

    Summary: SDS PAGE has been carried out on dermatophytes from two geographic locations. Differences were clearly observed between the species Microsporum canis, M. gypseum, M. equinum, M. nanum, Trichophyton mentagrophytes, T. erinacei and T. rubrum. More subtle variations were also apparent within the species M. canis.
