Summary: The ame of presented investigation was to determine the
activity of phosphoenolpyruvate carboxykinase in the liver of lactating
cows in vivo, in addition, to obtain the quantities of the liver tissue,
the biopsy method had to be devised. The results of the enzyme activity
proved to range between 0,87 and 3,4 Šmol/min/g tissue. The value
established are generally lower than those reported by other authors, which
could be due to the freezing of the tissue and keeping it frozen for longer
period of time and also to the different analytical method applied. It is
concluded that by using the presented biopsy and enzyme activity assay
methods, PEPCK can be successfully determined in the liver of dairy cows in
vivo.

Summary: The aim of the present Work was to determine the activity
of NADmalate dehydrogenase (MD) in vivo in the soluble fraction of the
liver tissue specimens obtained by biopsy during the first week of
lactation. The results, ranging from 101,6 to 243,2Šmol/g tissue/min,
showed slightly higher values of activity thanthose reported for the
healthy cows by other authors. It is concluded that by using the presented
biopsy and the enzyme assay methods, the activity of the mentioned enzyme
can be successfully determined in the liver of lactating cows in vivo. The
results are discussed in the light of the role of MD in the process of
oxaloacetate forming and trasporting pathways into the mitochondria and
cytosol of the cows hepatocyte, respectively.

Summary: In the present work the application of the method for the
urea analysis in milk but initially devised for the urea determination in
the blood, is presented. In order to prepare milk, the suitable medium for
this analysis the deproteinization and fat removing in the milk sample was
made ready. The analysis was always performed 24h. afther sample
collection. It is concluded that the method is accurate, simple and
convenient for the analysis of the urea in milk. Its simplicity makes its
application suitable for determining milk urea variations under practical
conditions in relation to production and the detection of nutritional and
metabolic disturbances in dairy cows.

Summary: In the paper the physiological basis of determining the
energy status of dairy cows by measuring the milk urea,fat and protein
contents is demonstrated,with the connection to the energy deficiency and
the incidence of respective health disturbances.The results of
investigation on the energetic status of the East-Friesian dairy cows and
the simenthal cows are presented.On the basis of the average values of
named parameters in milk,and also compared with the composition and way of
feeding,it is concluded that in both groups of animals there is pronounced
energy deficiency.

Summary: The biopsy method for obtaining liver tissue sample in
sufficient quantities for determination of liver enzyme activities on the
animals in regular production in farm is presented.The new instrument was
constructed with the diameter of cannula of 4,6 mm and modification of
standard technique was introduced.The sample weighing in average 0.52 g
were obtained from 49 cows of different ages,in comparation to 0,13 g in
the control group.The technique is convinient for the present purpose.

Summary: The low average milk urea (2,99ń0,62) and milk proteins
(2,71ń0,34%) concentrations showed energy deficit in great proportion of
animals in the experiment,most likely because of inadequate individual
supply of concentrate.The positive correlation coefficient (r=0,619)
between milk concentration and the phosphoenolpyruvate carboxikinase
activity (1,309ń0,56 Šmol/g/min) indicates that individuals with higher
milk urea content,approaching or entering the physiological range - meaning
less pronounced energy deficit,obviously displayed greater
phosphoenolpyruvate carboxykinase activity.
