Summary: In lactating dairy cows experiencing the metabolical
disorders and normal cows with good productive and reproductive features,
respectively, in the biopsates of liver tissue the activity of cytosolic
NAD-malate dehydrogenase and cytosolic phosphoenolpyruvate carboxykinase
are determinated in vivo. In addition, in the blood of experimental
animals the contet of total lipids, total and free cholesterol, glucosis,
free fatty acids, inorganic phosphorus and calcium are determinated. It is
expected that the obtained results will provide more detailed insight
into the gluconeogenic process in ruminants and give additional
explanations on the patogenesis of metabolic disorders. In the course of
the experiment, the new analytical methods giving insight into the
metabolic status of the animals are applied, which are at the same time
more practically applicable, namely the determination of urea and proteins
in the milk.
Research goals: The objective of the proposed investigation is to
examine the activity of extramitohondrial NAD-malat dehydrogenase (MD)
and phosphoenolpyruvate carboxikynase (PEPCK) in the hepatocytes of high
yielding cows in vivo, experiencing the metabolical disturbances (ketosis
or ruminal acidosis - conditions which include metabolical acidosis), and
in the normal cows with convenient productive and reproductive
characteristics respectively. The degree of metabolical disturbances will
be estimated by determinating the blood total lipids, total and free
cholesterol, glucose, free fatty acids, ketone bodies and calcium and
inorganic phosphorus. Since this field of ruminant phisiologyhas nor been
yet investigated in detail, the results of it would supplement existing
knoveledge on the activity of mentioned enzymes in the ruminants. In
addition, it is expected that the obtained results would faciliate the
insight into the extent of oxaloacetate formation from propionate in vivo
in the animals with disturbances, and on the importance of this part of
gluconeogenic pathway in the course of pathogenesis of metabolical
disturbances. It is also expected that the results of the enzyme activity
in liver could corelate with results of the measured parameters in blood
and milk, what could be applicated in the diagnostics as an index of the
development's degree of the metabolical disturbance. Finnaly, in the
course of the work, the new methodology of enzyme and metabolite
determinations will be gradually introduced.
