Title: Reversibility of the inhibitory effect of atrazine and lindane on cytosol 5 alpha-dihydrotestosterone-receptor complex formation in rat prostate

Summary: Both atrazine and lindane interfere with the physiologicalprocess of specific DHT-receptor complex formation in the ratprostate cytosol, but the effects are reversible. The process wasrestored after 22 days in young rats and after 14 days in adults,following the cessation of atrazine influence, while followinglindane influence, restoration was complete after 7 days for bothage groups. The atrazine or lindane applied doses can occure insome accidental situations or through continous use of pesticidesas field or storage exposure without adequate protection.

Summary: The effects of gama-1,2,3,4,5,6-hexachlorocyclohexane (lindane)and O,O-dimethyl-S-(1,2-dicarbetoxyethyl)phosphorodithioate(malathion) on testosterone metabolism in rat prostate tissue invitro was investigated. The influence of lindane on the formationof 5 alpha-androstane-17 beta-ol-3-one (5 alpha DHT)-receptorcomplex in the rat prostatic cytosol has also been examined.Lindane and malathion inhibited the formation of 5 alpha-DHT, andlindane caused a decrease in the formation of 5alpha-androstane-3,17-dione (androstanedione) in the rat prostatetissue. A mixture of lindane and malathion exerted a sinergisticeffect on testosterone metabolism in the rat prostate, asindicated by lower 5 alpha-DHT and higher 5 alpha-androstane-3alpha,17 beta-diol formation. The presence of lindanesignificantly inhibited 5 alpha-DHT-receptor compplex formationin the rat prostatic cytosol, and the inhibition was of a fullynon-competitive type.

Summary: To investigate a possible cytokinin activity, a novel syntheticcompound 1-(6-purinyl)-2,5-dimethylpyrrol (PDP) wasexperimentally tested by using the classical tobaccobioassay, and callus culture of Coleus blumei Benth. regardedas a suitable system for producing secondary metabolites(rosmarinic acid). The activity of PDP was compared tothat of kinetin (Kt). The results indicate that the selectedcompounds PDP has a stimulating effect on the growth of Coleusblumei Benth. callus and can be considered ascytokinin. The fluffy structure of Coleus blumei Benth. callushas shown some advantages in establishing cell suspensionculture.

Summary: The effects of subacute exposure to atrazine on body weight ovarian cycling, conception rate and the size of litters in Fisher strain rats were studied. An atrazine dose of 120 mg/kg body wt given p.o. daily for 7 days to both female and male rats caused statistically significant lose of body weight in both sexes during the period of treatment. After the cessation of treatment the body weight of female rats remained below control body weights at least for 2 weeks. In male rats the different in body weights narrowed with time and reached control level 5 days after the last dose. Atrazine increase weights of pituitary and prostate, disturbed ovarian cycling and decreased successful mating in the first week after treatment.

Summary: The influence of s-triazine compounds (atrazine, prometryne and deethylatrazine) on testosterone conversion and 5alpha-didydrotestosterone-receptor complex formation was studied in vitro and in vivo in the rat prostate. A marked in vitro influence of atrazine and prometryne (from 0.465 to 1.393 umol) and their mixtures (in total concentration, 0.928 umol) on 5alpha-dihydrotestosterone formation was detected. The inhibition of the enzymic activities responsible for testosterone conversion and steroid hormone-receptor complex formation was non-competitive and reversible, and s-triazine compounds act as antiandrogens.

Summary: The influence of the herbicide atrazine on the Fischer rat and their offspring was investigated. Six 120 mg doses of atrazine in paraffin oil per kg body weight were applied every 48 hours to the females in Group A. The females in the control Group C, received paraffin oil only. Atrazine did not affect estrus cycle, gestation and delivery, litter size and pup survival. Adult female offspring of Group A displayed increased activity in the activity cage and the male offspring avoided more electric shocks during the shuttle box acquisition trials. Group A offspring had shorter latency time during extinction trials than Group C offspring.
