Summary: The project comprises interdisciplinary research in the
field of chemistry, biochemistry, biology and immunology. The aim of the
project is: 1. The preparation of new compounds posessing
immunomodulating activity, with emphasis on new derivatives of
peptidoglycan monomer and new adamantyl peptides, including synthetic
modifications of peptide chain as well as the syntheses of adamantyl
peptides. Synthetic modifications of carbohydrate molecules and the use of
serum esterases for deprotection of carbohydrates will also be
investigated. 2. To test biological activity of prepared novel compounds
in experimental models in vivo and in vitro, including immunomodulating,
antitumor and antiviral activities. 3. The development of immunotests
(immunoradiometric and enzymeimmunoassays) for detection and quantitative
determination of biologicaly active compounds in biological material.
Research goals: The purpose of the proposed project has been the
study ofstructure-activiy relationship, particularly the effects
ofchemically well defined compounds on selected parameters ofimmune
system. Such interdisciplinary oriented project comprisesstudies in
different research fields and the results of thesestudies should provide
new insights into the possibility ofmodulation of biological activity and
immune response. 1. Native bacterial peptidoglycans are specially
interestingstructures possesing marked immunomodulating properties. In
thescope of the proposed project, new methods for the preparation
ofcompounds containing elements of peptidoglycan structure will
beinvestigated: a) modification of native peptidoglycan monomer(PGM) and
b) de novo syntheses of adamantylpeptides in whichadamantyl residue will
be coupled to peptidoglycan fragment. 2. The biological activity of
prepared novel compounds will betested in model systems in vivo and in
vitro. The effects oftested compounds will be estimated with regard to
possibleantitumor and antimetstatic activity, immunomodulating
propertiesand antiviral activity. 3. The preparations of appropriate
immunogens by coupling haptensto proteins will be studied, followed by
immunization of animalsin order to prepare specific antisera. New test
systems (radio-and enzymeimmunoassays) for the detection of biologically
activecompounds in biological material will be developed.
