Summary: The work on the project continues to be concerned with
ananalysis of the mechanisms and role of oncogene activation and growth
factors involvement in tumor growth. The research included the
characterization of some growth factors-like molecules and cell-cycle
regulatory molecules, including receptors, signal transducers,
transcription factors. Some of the signals originating from the receptors
need to enter the nucleus of the cells to activate particular genes. This
interaction is mediated by specific proteins (nuclear transcription
factors) that bind to region adjacent to the relevant genes. We have been
undertaking extensive studies on following oncogenes and growth factor:
c-myc, c-myb, c-H-ras, c-erbB-2/neu, EGF-R, PDGF, insulinoids,TGF alfa,
TGF beta. Our studies have indicated that a critical abnormality involved
in tumor development is acquistion by cells for an autocrine capacity to
produce their own growth factors. Details are published in listed papers.
Two major methodological improvements have been done: DNA amplifications
by polymerase chain reaction from cytological smears and
immunohistochemical localization of nuclear oncoproteins in paraffin
embedded tissues. Gene therapy for solid tumors using herpes simplex
thymidine kinase suicide gene has started. Details are published in listed
papers. All goals of the project have been realized. The project have had
significant impact on the development of molecular human genetic in
Republic of Croatia.

genes in human tumors. 7. the molecular aspects of tumor metastases. 8.
gene therapy of cancer. 9. cellular and molecular mechanisms of newly
synthesized anti-cancer drugs. 10. accumulation of multiple genetical
alterations in human tumors. 11. the role of cell surface molecules in
T-cell differentiation and function. 12. new approaches in molecular
diagnostics. 13. automatic oligonucleotide synthesis. All goals have been
realized in the period from 1991. to 1995.
