    Summary: Blood have been sampled by cardiac puncture from non anaestetizedsea bass from commercial fish farming facilities in Limski kanal(Center for Mariculture- MARIMIRNA, Rovinj). Comparison was madewith fish from the wild population. Several hematological(RBC,Hct,Hb,MCHC,MCV,MCH) and biochemical parameters (totallipids, proteins, cholesterol, blood sugar, albumin and globulin)have been measured. The obtained results indicated a significantseasonal variability related to the environmental seasonalvariations and particularly pertinent to the general metabolicactivity, sexual cycle and other biological function.

    Summary: The growt rate, feed conversion, hematological parameters andcomparative tissue composition were examined in three groups ofthe sea bass fed on different diets. When experiment was started,17 month-old fish had an average weight of 150-170g and thelenght of 23,5 - 24,6 cm. Two commercial (A and B) and oneexperimental (C) dry pellet diet were tested. Fish were sampledafter 73 and 130 days. The highest growth rate and final bodyweight after 73 days were observed in the fish fed on diet A.After prolonged feeding (130) days in all groups thegrowth rate indexes were reduced but among them the highestgrowth rate was maintained in group B. In general, a significantincrease of body fats was observed, and the highest accumulationwas in the liver and abdominal cavity.

    Summary: In north Adriatic like in other seas, the concentration of totalarsenic in foods of marine origin and in all marine animals isgenerally exceeding the concentration approved by the sanitaryfood quality regulation.     Summary: Three years after sinking of the "Brigitta Montanari" it wasnoticed that VCM started to leek from the wreck. This reportrepresents a part of complex reserach program that was organizedas requested by environmental protection authorities. VCM isknown as hepatotoxic and teratogenic agent which inducesangiosarcoma in liver of experimental animals and man. Thus ourinvestigations were focused to check the changes in activity ofliver specific enzymes (GOT,GPT and SDH) in representativebenthic fish caught by trawling nearby the wreckage. Twocommeercially important fish species were analyzed: europeanhake(Merluccius merluccius L.) and common pandora (Pagelluserythrinus).

    Summary: The present papers contribute to the identification andcharacterization of mussel (Mytilus galloprovincialis) esterases,so that these enzymes may be used as indicators of organophophateand carbamate poisoning. Esterase were detected in almost allmussel tissues but the highest activity was found inhepatopancreas. The existance of esterase isozymes was identifiedby starch gel electrophoresis. Combining the relative mobilitywith the substrate specificity and the sensitivity to theinhibitors, esterase from mussle hepatopancreas were classifiedas carboxylesterases and acetylsterases. In vitro studiesindicated that organophoshate and carbamate pesticides havedifferent effects on different esterase isozymes of mussel.

    Summary: This study was initiated to evaluate the changes in plasma enzymeactivity in marine fish after acute exposure to pollutants. Adultgrey mullets Mugil auratus were used as test organisms. Acuteliver injury was experimentaly induced with high carbontetrachloride and phenol doses. Lactate dehydrogenase (LDH),glutamate oxalacetate transaminase (GOT) and glutamate pyruvatetransaminase (GPT) were selected as relevant enzymes forevaluation of liver intoxication. Dose dependent increases of allthree enzymes in plasma were observed promptly after injection ofthe toxicant. The electrophoretic patterns of GOT from differentmullet tissues, and the organ/tissue distribution of both GOT andGPT suggested liver to be their origin.

    Summary: Besides glutamate oxaloacetate transaminase (GOT), glutamatepyruvate transaminase (GPT) and lactic dehydrogenase (LDH) wehave studied the possibility to use sorbitol dehydrogenase (SDH).glutamate dehydrogenase (GLDH) and alkaline phosphatase (AP) toexamine liver poisoning in grey mulet. Fish were treated withcarbon tetrachloride, phenol and cyanide. The increased activityof GOT,GPT, SDH and GLDH and the changes in concentration oftryglicerides in plasma can be used as positive indicators ofliver damage by poisoning. The changes in activity of AP, thedecreased concentration of plasma proteins, hemoglobin andhematocrit are discussed.

    Summary: Two aspartate aminotransferase isoenzymes were detected in the red muscle of grey mullet. One of them was purified to homogeneity by chromatography on hydroxylapatite, ion exchange columns, chromatofocusing and FPLC on the Mono Q column. The isolated enzyme is a dimeric protein of identical subunits with overall Mr about 65000, as judged by gel filtration and polyacrylamide gel electrophoresis. It has a three electrophoretically distinct subforms with isoelectric points of 8.50, 8.70 and 8.85. The Michaelis constants for L-aspartate, 2-oxoglutarate and oxaloacetate were estimated to be 0.30, 0.45 and 0.13 mM respectively, whereas Km for L-glutamate is 8.57 mM. According to its subcellular distribution, molecular, catalytic and inhibition properties isolated enzyme belongs to the mitochondrial form of aspartate aminotransferase.

    Summary: Toxicity of various polluting sdubstances was evaluated using seaurchin gametes and their early developing stages.THe changedsperm motility, fertilization, clevage and hatching rate, and thealtered DNA and echinochrome production were parameters of mainconcerns. The results about the evaluation of sperm motility bycentrifugation or by filtration of diluted sperm were notconsistent with all used pollutants. However, the best comparableresults were obtained by measurment of DNA and echinochromeproduction rate. The percentage of unfertilized eggs, thedecreased clevage rate and hatching time and the progressiveappearing of various morphological deformations, observed bymicroscopic examinations are the best actual evidence of thetoxic effects of the used substances.

    Summary: In this study the presence of cytosolic and mitochondrial aspartate aminotransferase isoenzymes in the red muscle of grey mullet, Mugil auratus Risso, was established. Aspartate aminotransferase isoenzymes were isolated from the homogenate of the red muscle and characterized. Separation of isoenzymes was achieved by hydroxylapatite column chromatography. Cytosolic aspartate aminotransferase isoenzyme with specific activity of 135 U/mg was prepared in electrophoretically homogenous form. It was a dimeric protein composed of identical subunits with overall Mr of about 65000. Isoelectric focusing revealed that cytosolic isoenzyme was an acidic protein composed of several active subforms. Mitochondrial aspartate aminotransferase isoenzyme was also prepared in a homogeneous state. Isolated isoenzyme had relative molecular weight of about 65000 and it consisted of two identical subunit like its cytosolic counterpart. For mitochondrial isoenzyme, three active subforms were identified by isoelectric focusing at pH 8.50, 8.70 and 8.85. The purified isoenzyme was unstable to freezing and above 40řC. By measuring its activity at different pH values, the enzyme stability was established in the pH range from 7.0 to 10.5. Both isoenzymes of aspartate aminotransferase obeyed Ping Pong Bi Bi kinetic mechanism of reaction. Isolation and characterization of aspartate aminotransferase isoenzymes from red muscle of grey mullet, demonstrated that it has the same isoenzymic system as other vertebrate species thus far studied. According to their catalytic properties and substrate affinities, aspartate aminotransferase isoenzymes from red muscle may be involved in the aspartate-malate shuttle as well as in the citric acid cycle.

    Summary: Aspartate aminotransferase isoenzymes have a central role in the amino acid metabolism and participate in maintaining redox balance via an aspartate-malate shuttle in aerobic tissues such as red muscle tissue. The aim of this study was to purify and characterize cytosolic isoenzyme of aspartate aminotransferase from the red muscle of grey mullet. Properties of the isolated isoenzyme will be compared with that of cytosolic isoenzymes from other vertebrate species and its possible physiological role will be discussed.
