Summary: DNA damage in marine invertebrates caused by model
pollution have been investigated. For this purpose methods for
determination of single strand breaks, alkali-labile sites and crosslinks,
without using ionizing radiation have been developed. Increased frequencies
of strand breaks and DNA crosslinks in natural mussel populations under the
influence of urban and industrial wastes, enable the classification of
sampling sites of Rovinj area according to genotoxic impact on mussel DNA.
The mechanism of DNA fragmentation in marine sponges involved in apoptosis,
and modulation of this process by environmental pollutants, have been
investigated, too. Expression of different genes involved in activation of
pollutants, DNA damage and repair, has been analysed, including CYP 1A1,
ubiquitin and heat-shock genes in fish, mussel and sponge. The results of
our investigations gave additional informations about molecular mechanisms
involved in activation and deactivation of contaminants, as well as DNA
damage and repair in marine organisms.
Research goals: The main goals of our investigations are
understanding of mechanisms involved in activation and deactivation of
pollutants in marine organisms, including induction of DNA damage and
repair and gene expression involved in those proccesses. Development of
methods for such measurements were neccessary. Results of these
investigations should make possible estimation of presence of genotoxic
pollutants in the sea as well as genotoxic risk assessment for marine
organisms.
