Summary: The presence of biogenic amines, histamine and
particularly5-hydroxytriptamine - serotonin - in vegetative and
generativeplant organs will be investigated. We shall try to identify
thephase of ripening in which the biosynthesis of serotonin occurs.We
shall try to determine the functions of biogenic amines intissues and
particularly hormone activity of serotonin and itsmetabolite
5-hydroxyindole-3-acetic acid by biotests and tissueculture in vitro. The
location of amine in tissue will bedetected by histochemical reactions. In
these investigations weshall use chromatographic (thin layer, HPLC),
electrophoretic,spectrophotometric, spectrophotofluorimetric
andspectrocolorimetric methods.
Research goals: 1. Investigation of the presence of serotonin, its
metabolites and other biogenic amines (tryptamine, histamine) in plants,
particularly in edible ones. 2. Detection of physiological function of
serotonin and 5-hydroxyindole-3-acetic acid in differentiation and plant
growth. 3. An attempt to inhibit serotonin transformation in
5-hydroxyindole-3-acetic acid by diamine oxidase activity. 4. Determination
of phase development of fruit when adaptive enzyme formation occurs in
serotonin synthesis. 5. Investigation of the presence of the above
specified substances in plants which they have not been identified to
date. Extraction will be carried out under cold conditions, concentration
under mild conditions, separation of substances on ion exchange column, by
paper and thin layer chromatography, electrophoresis, and detection by
spectrocolorimetric, spectrofluorimetric and spectrophotometric methods.
6. Amine in tissues will be located by histochemical methods. 7. Detection
of auxin activity of serotonin and 5-hydroxyindole-3-acetic acid will be
determined by Avena coleoptile biotest, on root of barley, leaf discs of
tobacco, by Allium test and tissue culture. Other information about the project.
