    Summary: In cells infected with robinia mosaic cucumovirus (RoMV) weestablished two basic types of crystalline virus inclusions:hollow crystals, of polyhedral and spherical to oval appearance,and ordinary polyhedral crystals. The former seem regularlypresent in the vacuole, and the latter in the cytoplasm. Theoptimal temperatures for development of RoMV crystallineinclusions were between 15-20 C at the period of longer days. Allforms of the crystals were more frequent in relatively youngercells in which more intense filling of the hollow crystals tookplace.

    Summary: On the basis of the references supplied, a short review of theviruses found in forest ecosystems, specially in forest treespecies, has been given. The spreading of viruses in forestecosystems and some data on interactions between viruses andother stress abiotic and biotic factors of forest trees areincluded in the article as well. Until now viruses in forestecosystems in Yugoslavia have not been investigated.

    Summary: Cucumber mosaic virus (CMV) was islated in Hungary and Yugoslaviafrom wild-growing Asclepias syriaca L. (common milkweed) plantsshowing mosaic, veinclearing, line-pattern or mosaic symptoms.Five to 35 per cent of the individuals were found to be affectedin different places. Three isolates of CMV (Asc/B76 and Asc/Hfrom Hungary as well as Asc/Y from Yugoslavia) were identified onthe basis of reactions of test plants and by agar-gel doublediffusion serological tests using antisera to CMV. The isolateAsc/B76 was also characterized by host range, serologicalcomparison with the To and D serotypes of CMV as well as byparticle morphology, aphid transmission, cross protection testsand physical properties in plant extracts. Based on theseresults, this isolate was classified within the To serotype (Bpathotype) of CMV.

    Summary: An attempt was made to apply rocket immunoelectrophoresis (RIE)technique for determination of serological differentiation index(SDI) values between three British strains of turnip yellowmosaic virus (TYMV) based on the height of "rocket" precipitates.The experiments were conducted in 1% agar gel containingantiserum to Edinburgh (E) strain of the virus, the holes beingcharged separately with purified E and two other strains.Following experiments with the E strain in several subsequenttwofold dilutions, a calibration graph showing linearproportionality was drawn in a coordinate system determined bysteps of virus dilutions and logarithm of "rockets" heights.By interpolation procedure SDIs of two heterologous strainsagainst E were found fairly comparable with SDI values obtainedby double radial immunodiffusion technique.

    Summary: The degree of adsorption of tobacco mosaic virus (TMV) particlesand turnip yellow mosaic virus (TYMV) to two types of soil (humicand sandy soil) was established by laboratory tests. Both typesof soil adsorbe in high percentage TMV (c.95%) as well as TYMV(nearly 100%). Infectivity of the humic soil which contained theadsorbed TMV particles was completely reduced.

    Summary: A pronounced yellow discoloration on leaves together with someleaf malformations and plant stunting were observed on hybridBrassica Perko PVH grown in north-eastern Croatia. On the basisof serological experiments, host range, symptoms in test plantsand cell alterations, the casual agent was identified as anisolate (P isolate) of turnip yellow mosaic virus (TYMV).Serologically and symptomatologically the isolate was closelyrelated to the Edinburgh isolate of TYMV, i.e. it belonged tostrain 1 of the virus. In several fields in the test region thevirus was found in nearly 100% of the plant specimens. Itsconcentration in leaf and fleshy root tissue of Perko PVH fieldplants was somewhat higher than 1 mg/g. After passage trough therabbit alimentary tract, the P isolate was inactivated. However,it was serologically demonstrated in the faeces.

    Summary: It has been found by ELISA-test that soybean mosaic virus (SMV)occurs in equal concentrations in all parts of embryo of seedproduced in infected soybean cultivars NS-9 and NS-16 which weregrown in southeastern part of Yugoslavia (the province ofKosovo). The correlation between percentage of seed coat mottlingsymptoms and SMV infection was significant in both cultivars.

    Summary: Mild mosaic, chlorotic spotting, stunting and mild distortionsymptoms were observed on leaves of Lagenaria siceraria (syn.: L.vulgaris) convar. clavatina in the vicinty of Zagreb, Yugoslavia.From the infected plants a virus was isolated and identified ascucumber mosaic virus (CMV) based on test plant reactions, crossprotection, serology, virus stability in sap, aphid and seedtransmissibility, and particle morphology. The virus isolate isdesignated CMV-LY.     Summary: An attempt was made to test the applicability of the singleradial immunodiffusion method for determination of serologicaldifferentiation index (SDI) in plan viruses, based on acomparison of the terminal size of the ring-shaped immuneprecipitates. Rabbit polyclonal antisera against Britishisolates/strains "Edinburgh" (E), "Northumberland" (N) and S ofturnip yellow mosaic virus (TYMV) and the virus strain E wereused. From reactions of heterologous antisera SDIs betweenstrains E and S and between E and N could be read from the graphdirectly. They were determined in decimal fractions (0,9 and 3,3,respectively), which is more precise than in commonly performedSDI determinations (SDI expressed in whole numbers) by precipitintests in free liqid or double radial immunodiffusion.

    Summary: Cucumber mosaic virus (CMV), previously isolated from an unknowncultivar of autumn garlic (Allium sativum L. subsp. sagittatumKuzn.), was transmitted to garlic cv. Moravan already infected bylatent filamentous viruses. Transmission of CMV was achieved byMyzus persicae and only from infected Nicotiana megalosiphon, N.glutinosa and Chenopodium quinoa but not from the garlic. Theinfected garlic plants growing in the open showed sporadicchlorotic streaks on leaves and flower stalk. The yield of cloveswas markedly reduced, i.e. 54 and 35%, in comparison to theplants which held latent viruses only. A rare presence of CMV ingarlic and other cultivated Allium species is dicussed.

    Summary: We describe the use of glycerol to reduce aggregation of tobaccostreak ilarvirus (TSV) particles caused by formaldehyde (FA)fixation. After treatment of partially purified TSV preparationswith 1% FA in the presence of 10% glycerol double radialimmunodiffusion tests revealed the presence of two to four timesmore reacting virus than in preparations treated with 1% FA only.Consequently, the method provides higher quality preparationsintended for different immunochemical purposes and biophysicaland biochemical analyses. We expect it to be useful in fixationof viruses which tend to aggregate in the presence of FA.

    Summary: Prominent virus-like ringspot symptoms were observed on theleaves of Dracaena surculosa Lindl. Attempts to transmit a casualvirus by mechanical inoculation on some conventional test plantsremained unsuccessful. However, leaf squash preparation revealedfilamentous virus particles, as well as structures which could beparts of "pinwheel" inclusions. dsRNA, correspondingapproximately to the replicative form of RNA of potyviruses, wasisolated from symptoms bearing leaves. The results obtainedsuggest that the ringspot symptoms on D. surculosa could beprovoked by a potyvirus infection.

    Summary: An aphidborne filamentous virus was isolated from themonocotyledon Sternbergia lutea with chloritic strips on theleaves. The virus provoked cytoplasmic cylindrical inclusions ininfected tissue suggesting a potyvirus infection. No virus seemsto have been recorded in this monocotyledonous plant up to now.

    Summary: Pelargonium line pattern virus (PLPV), a so far ungrouped virusisolated from Pelargonium zonale, provokes intracellular changesvery similar to those of some dianthoviruses. We found that thevirus sedimented as a single component, contained a singlepolypeptide species with an Mr of 38000 and a RNA molecule of1.2x10on6 daltons. As PLPV was proved to be characterized withmonopartite genome, it is impossible to propose the affiliationof this virus species to dianthovirus group, despite very similarcytopathic effect.

    Summary: From different genotypes of soybean included in the breedingprogram of the Faculty of Agricultural Science in Zagreb,seventeen isolates of soybean mosaic potyvirus (SMV) wereobtained. Their identity was established by host radge study,morphology of virus particles, and serology. the virulence ofobtained isolates was established according to the severity ofthe reactions they induced in specific soybean differentialcultivars. The presence of SMV isolates of very low virulence indomestic and introduced cultivars indicates that these cultivarspossess a very low resistance to SMV and that the breedingstrategy must be adjusted to put more emphasis on resistance toSMV.

    Summary: Several approaches for detectiuon and diagnosis of plant virusescan be used. Some new methods, as well as simplificationsof some already known, have been introduced. One of themost important methods is the isolation and analysis ofreplicative double - stranded RNA (dsRNA) from, virus infectedplants which became consistently present in many attempts ofdaignosis and all recent attpempts of plant viruscharacterization. The procedure is accomplished by extraction oftotal nucleic acids from infected plan tissue using organicsolvents, further purification of dsRNA by means ofphosphocelulose column cromatography and electrophoretic analysisobtained dsRNA. The electrophoretic bands, resulting from suchexperiments, represents replicative form dsRNA of roughly twicethe molecular weight of the encapsidated genomic single strandedRNA. Frequently, additional minor dsRNAs of faster mibility wereobtained. At this moment the origin of the latter is obscure, buttheir presence in preparations from infecteds plants enhances thediagnostic value of the technique. The less prominent dsRNAs areconstant and enable the differentiation of viruses within agroup. Sometimes, even strains of some viruses could be resolved.The replicative RNA is very resistant to enzymatic degradation.As it is not normally present in healthy plants and aould beisolated from low amount of tissue, the method has beenrecommended as rapid and reliable for diagnosting diseases andidentifying pathogens.

Walp.), growing as a wild planton the Dalmatian island of Dugi and showing dark green and/oryellow mosaic and vein banding, a filamentous virus has beenisolated and studied.     Summary: Optimal parameters for purification of dsRNA from aqueous phenol-extract of virus infected plant tissue were given. Chromatography was performed by directly adding CF-11 cellulose to the extract of total nucleic acid, and collecting it by centrifugation after stirring the mixture (adsorption step). Washing steps and elution were accomplished in the same way. Very good result is available in less than one hour, even when the plant extract is viscous, making the standard column chromatography difficult.

    Summary: Double-stranded RNA replicative forms of genomic and subgenomic viral ssRNA and additional dsRNAs were isolated by chromatography on CF-11 cellulose from phenol-extracts of plants infected with turnip yellow mosaic and from phenol-extracts of plants infected with turnip yellow mosaic and belladonna mottle virus. Obtained agarose gel electrophorograms were characteristic and sufficiently similar to be diagnostic for tymoviruses, but also clearly distinct for each virus. Small differences were detected between virus strains and from different hosts infected with single virus isolate.

    Summary: Viruses contribute to the phenomenon of forest decline, either bytheir presence in living plants or by being deposited in soil andwater as a reservoir of infections. The present report providesevidence on contamination of forest soil and draining (brook andditch) water with tobacco necrosis virus (TNV) and with twotobamoviruses, i.e. tobacco mosaic (TMV) and tomato mosaic (ToMV)viruses, respectively, in a part of central Croatia. For virustrapping and isolating from soil, soil/root samples, bait plantsand virus test plants were used. Virus isolation from waterfollowed the inoculation of virus test plants with concetratedsuspensions of high - or low speed pelleted water samples. It wasfound that forest soil was heavily contaminated (24 out of 49soil samples) with TNV. Tobamoviruses (common TMV and commonToMV) were contained in a comparatively high titre in forestbrook (TMV and ToMV) and forest ditch (TMV) water. The criteriaof symptomatology, particle morphology, cytopathology andserology were applied in virus identification.

    Summary: Parellel light and electron microscopy investigation revealed genesis of the hollow crystals of robinia strain of peanut stunt cucumovirus (PSV-RoM), and by cucumoviruses in general. It was also found that small tonoplast vesicles (probably places of viral RNA synthesis) are characteristics cytopathic changes for PSV-RoM, and consequently for all known cucumoviruses. Occurence of dark stained endoplasmic reticulum implies to changed activity of that cell organell.

    Summary: The quantitative dominant flavonoid was isolated from flowers of endemic species Centaurea rupestris L. By means of spectral analysis (UV, mass NMR) it was chemicaly identified as 7 O beta D glycopyranoside which aglycone is 3,4',5,6,7-pentahydroxy-3-metoxyflavon. It was established the significant antiviral activity of mentioned glycopyranoside against tomato bushy stunt virus (TBSV). The mechanism of this action was explained. The antifungal and antibacterial activity of isolated flavonoid was established, too.

    Summary: Tomato fields in the southern coastal part of Croatia were stricken by lethal necrosis on such a scale it endangered tomato production in the area. Cucumber mosaic cucumovirus (CMV) infection was proved by serology. Low-molecular weight RNA band was detected in dsRNA and TNA samples from naturally infected tomato fruits. The satellite nature of this band was proved by molecular hybridization. This is the first report of CMV with a satellite RNA in Croatia.
