Summary: Gangliosides are involved in modulation of cell
membrane receptors, especially those for "growth factors", their content
and structure alteration might be connected with the cell's malignant
transformation, and therefore their investigation might enrich our
knowledge and further understandings on pathogenesis of anterior pituitary
tumors of endocrinologists as well as oncologists, that might certainly
bring us to more successfull treatment of these lesions. The aim of our
investigation is to establish the quantitative and qualitative
characteristics of ganglioside fractions in serum and tumorous tissues as
well, to delineate them according to the kind of secreting hormones, and
their pathohistologic features, so in that way to determine pathognomonic
"markers" for the reliable diagnosis and differential diagnosis of existing
lesions. By means of monoclonal cell lines from the tumor tissue cultures
we intend to define the characteristic ganglioside fractions, and more
thoroughly the secreting hormones as well. By pretreatment with the
available "growth factors", stimulating and inhibiting ones, and
hypothalamic releasing hormones, we hope to identify their mutual
interconnection in the oncogenetic process of pituitary tumors. Such
understandings might disclose the creation of critical antibodies for the
diagnosis by means of specific "markers", as well as for the eventual
nonaggressive therapy.
Research goals: The general aim of our investigation is to get
acquainted further with structural and functional features of gangliosides
in the whole normal pituitary and in anterior pituitary tumors, as one of
the possible "growth factors" or a receptor for "growth factors" in the
malignent transformation of cells. The first and unique data in the
literature on the elevated serum gangliosides levels in acromegaly, as
well as the first and unique analysis of tissue gangliosides in the
anterior and posterior lobes of the normal subjects and patients with
hormonally active pituitary adenomas, are the result of our studies. By
defining the specific "markers" for various anterior pituitary adenomas,
we will be able to discover the recurring lesions of the unsuccessfully
operated patients at earliest possible term. One of the therapeutical
approaches in the treatment of the anterior pituitary tumors could be the
inhibition of autocrine and paracrine actions of "growth factors" by means
of antibodies or synthetic peptides, i.e., by means of directing the
toxin-antibodies onto the cell surface "growth factors".
