    Summary: The surface glycoprotein S (spike) of coronaviruses in a major determinant for virulence and displays great phenotypic variability among different strains and variants. Here we addressed the question, whether this variability is reflected by a heterogeneous quasispecies-distribution of mutated RNA-molecules within the infected organ. As a model, we analyzed the variability of S-gene sequence directly in brain tissue from rats persistently infected with murine coronavirus. Our results indicate, that in vivo-at least under the constraints of a persistent infection-the S-protein sequence appears to be relatively conserved.

    Summary: Much evidence shows that somatostatin plays a significant role inthe circuit that exists between the neuroendocrine and immunesystem. Its mostly immunosuppressive effects may be related toits capacity: 1) to modulate the release and action of variousneurotransmitters, hormones and cytokines which affect thelymphatic functions; 2) to act directly on receptors oflymphocytes and other cells participating in immune response,and/or 3) to change the other events related to the function oflymphatic organs such as blood flow, vascular permeability orlymphocyte traffic. Its widespread localization andmultispecifity of action emphasize its importance for themaintenance of neuro-endocrino-immune homeostasis.

    Summary: Juvenile DA and F1 hybrid rats (AOxDA) were immunized with bovinebrain white matter in complete Freund's adjuvant to produce therelapsing form of experimental allergic encephalomyelitis(R-EAE). Recovery from actively induced disease resulted in adifferent rate of susceptibility to active reinduction of diseasein these high susceptible rats. Hydrids F1 (AOxDA) developedrelative resistance, while DA rats were completely resistant toactive reinduction of disease. The treatment withcyclophosphamide (20 mg/kg), administered 2 days beforereimmunizaion, inhibited the resistance to EAE reinductionobserved in DA rats, and increased the susceptibility to EAEreinduction in F1 hybdids. Rats which were splenectomized 12 daysbefore the initial immunization were higly susceptible to activeEAE reinduction. However, when splenectomy was performed 4 daysafter encephalitogenic challenge (i.e. during the induction phaseof disease), significantly lower susceptibility to active EAEreinduction was observed. These results suggest that low dosecyclophosphamide sensitive (suppressor) cells and spleendependent suppressor activity could directly mediate themechanisms of acquiring resistance to active disease reinduction.

    Summary: Experimental allergic encephalomyelitis (EAE) was induced in ratsof various genotypes by active immunization with anencephalitogenic bovine brain extract. Chronic relapsing clinicaldisease was noted in juvenile F1 progeny of AO males and DAfemales in a high percentage of animals (65-75%), while in DAstrain it was only 20-25%. Here, a summary ofclinicopathological, genetical and immunological analyses isdecribed. The results corfirm: 1) the close similarities inmorphology of CNS lesions in CR-EAE and multiple sclerosis, 2)the essential role of cell-mediated immunity in the pathogenesisof EAE, 3) the fact that EAE susceptibility is a genetic traitand 4) the existence of a defective systemic immune regulation inCR-EAE.

    Summary: The effects of pharmacological pinealectomy (Px) on liverregeneration and lymphatic morphostasis were studied in ratstreated by evening injections of propranolol. The data have shownthat Px might inhibit the early phase of liver growth and expression of class II antigens in regenerating tissue. Theseeffects were followed by marked changes in the spleen (in theproportion of CD4, CD8, OX-33, OX-42, Mac-1 and Mac-3+ cells).Some of these effects might be reversed by melatonin.

    Summary: In the present study were studied the biological activities ofthe immature forms of two peptide, isolated from activatedneutrophils-proBac5 and proBac7. The data have shown that proBac7is a monocyte-selective chemoattractant, while proBac5 is anefficient inhibitor of cathepsin L, a cysteine proteinase, whichcontributes to local tissue injury.

    Summary: The effects of long acting aomatostatin analog SMS 201-995 were examined in vivo on: Lymphoid morphostasis and functional reactivity of cells obtained from SMS treated donors, on humoral, and cellular type of immunity, and in vitro on blastic transformation of lymphocytes stimulated by activators of different transmembrane pathways (CD2 by PHA and CD#/TRC by anti-CD3 monoclonal antibody and by allogeneic cells) and on growth and secretory activity of several hybridoma cell lines. The data have shown that SMS in vivo decreases the proportion of CD4,CD5 and Ig+ cells in spleen. The reactivity of these cells to Con A was suppressed, but their spontaneous blastic transformation was increased. SMS suppressed also the PFC generation and proliferation of cells in popliteal lymph nodes during the local host versus graft reaction. The data obtained in vitro revealed that SMS may inhibit only CD2-induced blastogenesis (in early and late interval after the use of PHA). SMS inhibited also the spontaneous growth and/or secretion of antibodies in some hybridoma cell lines.

    Title: Regeneration of hepatic and lymphatic tissue after the depletion of T lymphocyte subpopulations

    Summary: The aim of this work was to establish: 1) the features of thecompensatory liver growth, after partial hepatectomy (pHx), 2)the effects of in vivo depletion of CD4, CD8 and CD4+CD8lymphocytes on the processes of liver regeneration and 3) theeffects of pHx on the spontaneous repopulation of lymphatictissue in depleted animals. The data have shown that pHxincreases the expression of MHC class II antigens in theregenerating liver and induces a marked changes in lymphaticmorphostasis (phenotypic changes in spleen, hiperplasia in bonemarrow, appearance of autoantibodies in sera).It seems that someof Hx-induced cells perform the morfogenetic function, becauseafter the depletion of CD8+ lymphocytes an increase in hepaticRNA and proteins was noticed, while after the depletion ofCD4+CD8+ lymphocytes an earlier accumulation of hepatic proteinsoccured. During the time when the liver regeneration has stoped,an increase of CD8+ lymphocytes in spleen, and in titer ofanti-DNA antibodies in sera were noticed. Taked together, thedata point to the role of lymphatic cells in the control ofnormal growth and to the hepatic origin of substances thatcontrol the lymphatic morphostasis.

    Summary: In this study, the biological activities of the proforms ofbactenecins, which do not exhibit antimicrobial effects have beeninvestigated with polypeptides purified by ion-exchange andpreparative reverse phase liquid chromatography. It was chownthat proBa7 is a monocyte-selective chemoattractant. The matureforms and proBay5 did not show this activity. In vitro, theprobactenecins did not affect the blastic transformation oflymphocytes. Enzimatic analysis has shown that proBac5efficiently inhibits the activity of cathepsin L.

    Summary: The effects of somatostatin (SMS 201-995) were investigated1) in vivo: on humoral and cellular immunity and morphostasis oflymphatic system and 2) in vitro: on blastic transformation oflymphocytes stimulated by activators of different trasductionpathways and on phenotypic profile of cultured cells. The datahave shown that SMS has a marked immunosuppressive activity. Itsantimitotic effects on blastic transformation of humanlymphocytes, stimulated by anti-CD3 monoclonal antibodies andPMA, however, did not affect the expression of early activationmarkers (IL-2R), nor some tyrozin protein kinase-relatedevents (down regulation of CD4 molecule), indicating that itsintracellular action goes through the others pathways.

    Summary: In mice immunosuppressed by halothane anaesthesia and operativestress the effects of peptidoglycan monomer, its analogues andindomethacin were investigated. The data have shown that PGM, andespecially its N-acil and methal complexes may correct thehumoral type of immunity in these mice. The effect was followedby hyperplasia of bone marrow and an increase of hepaticproteins. Similar immunocorrection was obtained byindomethacin.

    Summary: We previously observed that PGM, linked with zinc (PGM-Zn, "Pliva"-Zagreb) possesses the immunocorrective effects in some experimentaly induced or naturaly occuring immunodeficient states.To obtain information about the mechanism of the action of PGM-Zn, in this work we examined the effects of PGM-Zn in SRBC sensitized mice, which were in vivo depleted of CD4+ and/or CD8+ cells (by analysis of PFC generation, titar of antigen specific and unspecific antibodies, class II expression in the liver, phagocytic activity of peritoneal macrophages). The results revealed that PGM-Zn, in spite of absence of CD4+ or CD8+ lymphocytes, can stimulate the humoral immunity. After single depletion PFC production was enhanced in both primary and secondary response. In the case of double depletion only secondary response was increased. The reaction was followed by remarcable increase of antigen specific and nonspecific IgM titer in sera in primary reaction, and IgG and IgG1 titer in secondary reaction, and great expression of class II antigens in the liver. PGM-Zn stimulated also the phagocytic activity of peritoneal macrophages, which in coculture with syngeneic splenocytes, stimulated with ConA showed suppressive activities. Additional data, obtained in vitro, showed that PGM-Zn has the ability to delay the early process of internalization of CD4 molecule in human lymphocytes, stimulated by anti CD3+PMA, and accelerate the appearance of IL-2R molecules on the surface of spontaneously growing cells.
