Summary: Investigation of molecular basis of metabolic disorders was undertaken in order to develop and test the experimental model on the example of investiga- tion of genetic factors of atherosclerosis and lipoprotein metabolism on similar disorders. Modern methods of molecular biotechnology have been established and probes synthesized. The experience and skills were presented to the participants of postcongress course "Methods of molecular biotechnol- ogy in clinical laboratory", also in the form of detailed instructions published in a textbook. The proven successful model of metabolic disorders investiga- tion was applied with certain modification in the research of dyslipoproteinamia, chronic myeloid leukemia, HLA-C system, cystic fibrosis, ganglioside in hereditary neuropa- thies, endemic nephropathy, MCAD gene, biotinidase and bone metabolic diseases. The investigation included lipoprotein characterisation in coronary heart disease, myocardial infarction, postmenopause with hormone replacement therapy. More subjects were included in the investigation of apoE polymorphism in chronic dementia and Alzheimer's disease. We are collaborating in the estimation of reference values for apoE within the multicenter project. The applied methods were adjusted according to the ISO standards recommendations.

Research goals: investigation of molecular basis of metabolic disorders,investigation of genetic factors of atherosclerosis, study genetic variation of Lp(a) and apo(a) and plasminogen, establish polyclonal and monoclonal antibodies anti Lp(a) and anti apo(a), reproduce and determine sequence ofspecific apo(a) region in order to obtain probes for genetic screening, investigate genetic variations of apo A, apo B and apo E and their influence on dyslipoproteinaemias, study the role of MCM and LCAT in lipoprotein catabolism, analyze molecular regulation of binding kinetics of normaland structurally modified lipoproteins to specific receptors, investigate frequency of A985G mutation in MCAD gene, establish bcr/abl translocation in chronic myeloid leukemia, analyze genetic basis of cystic fibrosis, study the role of ganglioside in hereditary neuropathies, investigate etiology of endemic nephropathy, study pathophysiology of metabolic bone diseases, analyse lipoprotein characteristics in coronary heart disease, myocardial infarction, postmenopause (with hormone replacement therapy), investigate apoE polymorphism in chronic dementia and Alzheimer's disease, contribute in the multicenter estimation of apoE reference values
