Summary: The activity of peptidoglycan monomer and its
pharmacokinetics will be studied. Among biological activities, the
influence of peptidoglycan monomer on humoral and cell activity;
immunostimulation characteristics and other effects on the organism will be
studied. By toxicological research acute and chronic toxicity, reproduction
toxicity and genotoxicity of peptidoglycan monomer will be examined.
Pharmacokinetical studies will include the experiment with labelled
peptidoglycan monomer and pharmacokinetics with application of suitable
analytical method for measuring in biological material. In preliminary
research, the suitable computer pharmacokinetic model for calculation of
pharmacokinetic parameters from experimental data will be established.
Research goals: The goal of further studies is to clear up the
activity mechanism, to find out the additional activities and to clear up
pharmacokinetics in the animal model. Concerning the activity study,
immunostimulation characteristics will be examined in conditions of
experimentally produced immunodepression, similar to immunodepression that
can appear on clinics after the general endotracheal anaesthesia. Activity
against the viral infection will be examined on a submersed human leukocyte
culture concerning the ability of peptidoglycan monomer to provoke
interferon and other lymphokines production. The main goal of the
pharmacokinetical research is to achieve the conditions (if
pharmacodinamical and toxicological results are positive) for
pharmacological opinion, necessary for approval license to start the first
phase of clinical trials. At the same time the efforts to find out the
appropriate analytical method for peptidoglycan monomer measuring in
suitable biological materials will be done. Besides, the several
pharmacokinetical models will be set up and check out, verified by
application on pharmacokinetics of well-known drugs in animal and human
models. Tissue distribution of drugs, specially of peptidoglycan monomer
and its kinetics will be of special interest. Other information about the project.
