Summary: Sorbitol is an intermediate in the poliol pathway that convertes glucose to fructose. Intracelullar accumaulation of sorbitol has been implicated in the etiology of long-term diabetic complication. Erythrocyte sorbitol content and plasma glucose level were measured, and sorbitol-glucose ratios of patients with IDDM, NIDDM and healthy control subjects were calculated. Erythrocite sorbitol levels were markedly elevated in diabetic patients in comparison with normal subjects. There were no significant differences in sorbitol-glucosa ratios between those two groups, but the ratios were markedly lower in diabetic patients with complications. We conclude thatn red cell sorbitol level increases in relation to plazma glucose levels, but the cause of diabetic microangiopathy can not be explained only by overactivity of the polil pathway resulting from hyperglycemia and can probably be attributed to the interaction of as yet unknown multiple metabolic factors

Research goals: The goal of investigation is devided in two directions: clinical and analitic. A)In clinical use investigetion includes exploration of carbochidrate methabolism in insulin dependent and independent diabetics, and also better prevetion, diagnostic and treatment of chronic diabetic complications. Influence of the sorbitol level on developement of chronical complications in diabetics and also reversibility of complications will be followed. B)In analitic investigation we compere two different methods of erythrocyte separation that are used in detection of sorbitol. We also try to compare those two methods with the third one which has still been developing.
