Summary: As a model for the induction of diarrhea in weaned pigs, the F4+ ETEC parental strain M1823 or oxytetracycline/nalidixic acid resistant mutant M1823B in F4 susceptible pigs were used. The model includes feeding a diet containing 240ml of Colimix vaccine per 1 tone of creep or starter feed. Diarrhea developed in F4-susceptible pigs inoculated with M1823 (8 of 9 pigs) or M1823B (4 of 7 pigs) and 10 or 4 diarrheic principals died by day 21 after inoculation, respectively. All but one F4-susceptible controls remained clinically normal. The F4 resistant principals and controls gained weight faster (P<0.01;<0.05, respectively) than did their F4 susceptible counterparts. The Colimix-treated F4 susceptible or resistant controls gained weight at similar rates during 3-week post vaccination period. During this period,the F4 resistant principals gained weight better (P<0.01) than their F4 susceptible conterparts. The ELISA titers for anti-F4 antibody in the sera and intestinal secretions from pigs inoculated with M1823 or M1823B were much higher (P<0.001;<0.01, respectively) than those from the respective controls.

Summary: Nineteen of 22 neonates (86%), 17 of 20 weaners (85%), and 26 of 39 adults (76%) were classified identically as F4 (K88)receptor-positive or negative by the ELISA. The ELISA with feces from adult swine was found to be almost equally specific (87%) as that with feces from neonatal (90%) and weaned (91%) pigs. However, the sensitivity of the assay was low (38%), indicating that fecal samples from adults contained less receptor-material than necessary for comparable phenotyping. The receptor-positive brush borders from neonates and weaners reacted significantly better (P<0.02, <0.001 respectively) with purified F4 (K88) antigen that did those from adults. There was good agreement between the average ELISA values for feces from infant and adult swine regrdless the source of coating antigen applied. With this assay we can determine F4 (K88) phenotypes of infant swine using easily-collected fecal samples rather than isolated brush borders.

Summary: The modulating effect of Brevibacterium divaricatum cell wall derivatives, i.e. peptidoglycan monomer (PGM) and peptidoglycan polymer (PGP) on phytohemagglutinin (PHA)-induced responses of peripheral blood lymphocytes (PBL) and spleen lymphocytes (SPL) from neonatal pigs farrowed to gilts or sows was tested. Both, PBL from progeny of sows and SPL from progeny of gilts responded significantly lower (P<0.01) when PGM (14mg/kg) was given by intraperitoneal (IP) route at Day 0 and Day 7 after farrowing as compared to the respective controls. When the same amount of PGM was given i.p. at Day 7 and Day 14, the responses of PBL and SPL from offspring of gilts were much higher (P<0.01) than those of nontreated controls. The treatment with PGP (2.5mg/kg) by the same route at Day 7 and Day 14 strongly stimulated (P<0.01) the responses of PBL and SPL of pigs originating from litters of either gilts or sows, respectively.

Summary: Four porcine strains of Escherichia coli were examined for their effects on the small intestine of 4-week-old weaned pigs infected orogastrically. The strains used experimentally were: strain 1467 (adhesin negative, non-toxigenic); strain 2407 and 1466 (adhesin positive, non-toxigenic); derived by genetical engineering from strain 1467 and containing a wild type plasmid and a recombinant plasmid, respectively, encoding the F4 antigen (adhesin); and strain M1823 (adhesin positive, toxigenic). In addition, 2-week-old pigs that died from natural colibacillosis associated with two strains ("Ihan 1 and 2"; adhesin positive, toxigenic) were examined. Strain M1823 and the Ihan strains produced moderate and marked lesions, respectively. Strain 1467 did not cause mucosal damage or an inflammatory response. Strains 1466 and 2407 caused a mild to moderate leukocyte (mononuclear and polymorphonuclear) infiltration in the jejunal (but not ileal) lamina propria. However, unlike strain 1466, strain 2407 did not cause damage to the small intestinal mucosa and should be futher studied as a potential oral vaccine strain for post-weaning E. coli diarrhea.

Summary: As results of the 1st International Swine CD Workshop, four monoclonal antibodies (mAbs) (numbers 002, 054, 118 and 127) clustered closely to the internal standard anti-porcine CD4 mAb, 74-12-4. To futher characterize the relationship between these mAbs, they were used in flow cytometry to inhibit binding of 74-12-4 to porcine lymphocytes. mAbs #002 (74-12-4) and #054 (PT90), completely inhibited, while mAb #127 (10.2H2) and #118 (b38c6) partially inhibited (57% and 77% respectively), the binding of 74-12-4. Futhermore, none of the mAb bound to a 74-12-4 negative strain of pigs. We conclude that the four mAbs bind to the same, or a closely related, epitope on the porcine lymphocytes.

Summary: Among all monoclonal antibodies (mAbs) analyzed in the 1st porcine CD workshop, six mAbs showed reactivity to the porcine CD5 antigen (workshop mAbs 067, 068, 069, 070, 071 and 119). Because of lack of immunoprecipitation studies for five (067, 068, 069, 070 and 071) out of the six mAbs, only one mAb (119) could be definitely characterized as mAb against the monomeric 63 kDa porcine CD5 antigen. The five other mAbs included in this cluster are recognized by an identical labelling pattern in FCM and competition of the CD5 epitope recognized by mAb 119. These mAbsy were allocated to the wCD5 subcluster.

Summary: Amongst the monoclonal antibodies (mAbs) submitted to the 1st International Swine CD Workshop, two mAbs (numbers 055 and 120) could be identified to recognize the porcine CD6 analogue. Both mAbs seemed to be highly T-cells specific and showed neither reactivity with cells of the myeloic lineage nor with B lymphocytes. The observed molecular mass of the antigen precipetated by mAb 120 of 110 kDa confirmed this classification. Without molecular analyses of the antigen recognized by mAb 055, but similar staining pattern in FCM compared with 120, mAb 055 was allocated to the wCD6 subcluster.

Summary: Among all mAb submitted to the first porcine CD workshop, based on FCM analyses six mAb could be identified to recognize the porcine CD8 analogue (workshop No. 004, 051, 052, 053, 108 and 109). In immunoprecipitation studies three mAb (Nos. 004, 108 and 109) recognized an antigen with an apparent molecular mass of about 35 kDa under reducting conditions and about 70 kDa under non-reducing conditions. The molecular masses of the antigens recognized by the three other mAb (Nos. 051, 052 and 053) are still unknown. Epitope analyses performed by blocking experiments led to the determination of two CD8 epitopes: CD8a and CD8b. CD8a is recognized by mAb Nos. 004, 051 and 052, and CD8b by Nos. 053, 108 and 109.

Summary: Among 54 mAbs determined to be reactive with porcine T lymphocytes and/or activation antigens, eight mAbs (workshop Nos. 005, 031, 080, 091, 092, 093, 094 and 110) derived from different laboratories grouped together in the T11 cluster and were ordered into the SWC1. One mAb (No. 111) which belong also in this group was lost during the workshop. The SWC1 antigen is a molecule expressed on the majority of leukocytes, resting T lymphocytes, monocytes and granulocytes, but not on B lymphocytes. On T lymphocytes it is down-regulated after activation. The molecular mass of the antigen is unknown. Epitope analyses revealed that seven out of nine mAbs recognized similar epitopes on SWC1 molecule.

Summary: The objective of this paper is neither to consider the secretion and transport of antibodies at mucosal surfaces of porcine gut nor to describe lymphocyte traffic and homing in the GALT compartments. Summary: Distinct pattern of CD antigen expression on T lymphocyte subsets and distribution of CD+ T cell in jejunal lamina propria (LP), Payer's patches (PP), and ileal mesenteric lymph node (MLN) have been correlated with diarrheal disease or local protection due to enterotoxigenic Escherichia coli (ETEC)/non-ETEC infection/vaccination in 4-week-old pigs. An inecrease of CD2a+, CD8a+ in LP, and SWC1a+ cells in MLN (P<0.05;<0.01, respectively) of pigs infected with ETEC strain M1823 was observed. Non-ETEC strain 2407 stimulated a strong proliferation of cytolitic CD8a+ cells in LP (P<0.01). pigs vaccinated with 2407 strain had more abbundant SWC1a+ and CD8a cells in MLN (p<0.05) or CD4a cells in PP (p<0.01), respectively. Numerous SWC1a cells were found in LP of villous epithelium, PP and paracortical/sinusoidal areas of MLN in M1823-treated pigs. In 2407-inoculated pigs marked infiltration/proliferation of CD8a+ cells were demonstrated in LP/PP and paracortex of MLN. Recombinant nontoxigenic strain 2407 seems to be a good source of protective (fimbrial) antigen for development of a safe and effective live oral vaccine against postweaning colibacillosis in swine.

Summary: As determined by parallel testing of brush broders, 62 (95%) out of 65 microscopic adhesion assay (MAA)-phenotyped pigs were classified identically on the basis of ELISA and MAA results. Although the ELISA with fecal samples was found to be less sensitive (80% to 90%), it was equally as specific (100%) as the assay with brush border samples to identify K88-resistent pigs consistently with the MAA. Moreover, screening of porcine feceses for K88 receptors by ELISA provides convenient and alternative means of identifying K88-adhesiveness in live pigs.

Summary: The effects of porcine non-enterotoxigenic E. coli (non-ETEC) strains 1466 and 2407 containing either wild-type or recombinant plasmids, encoding F4ac antigen were studied to establish their role in (1) causing mucosal damage of the gut, (2) inducing an inflammatory reaction of the gut-associated lymphoid tissue (GALT), i.e. in the small intestinal lamina propria (LP) and ileal mesenteric lymph node (MLN), and (3) stimulatig the protective immune responses mediated by CD+ T/B/leukocyte subsets in LP and MLN of experimentally infected weaned pigs. To differ from F4ac+ ETEC strain M1823, both non-ETEC strains induced only a mild swelling of jejunal/ileal mucosa and a moderate leukocyte infiltration in LP. An increase of CD1+ and SWC2+ cells was detected by single colour FCM among B and T lymphocytes, respectively, from LP and MLN of 2407-infected pigs. A strong proliferation of CD45+ cells was observed in LP/MLN of pigs infected with either 2407 or M1823 strain. The later strain activated CD8a+ cytolytic T cells in these tissues. The use of non-ETEC strain 2407 was associated with a marked proliferation of CD1+ and SWC2+ cells in both tested compartments of porcine GALT.
