    Summary: Of 32 pigs in which E.coli fimbriae were found 16 had F4 (K88) antigen (50%), 10 had F5 (K99) antigen (31.2%), 3 had F6 (987P) antigen (9.4%), and 3 had F4 and F6 antigens (9.4%) simultaneously. The remaining 3 isolates were nontypable with used antisera. Fifteen of 35 isolates examined for hemolysin production were hemolytic. Of these, 60% (9 of 15) were F4+ and 80% (12 of 15) originated from suckling pigs. Thirty-five per cent (7 of 20) of nonhemolytic isolates were F4+. Of 35 E. coli isolates 29 (83%) were sensitive to gentamycin, 18 (51%) to carbenicillin or trimethroprim, and 14 (40%) to cloramphenicol. Resistance to tetracyclin or sulfamethoxazole occurred most commonly (91% or 32 of 35), while a lack of any effect of lincomycin on bacterial cell growth was observed. Results of our study demonstrate that fimbriated E.coli strains are a major cause of enteritis in suckling and weaned pigs, reared under intensive conditions, and that in both age groups F4 was the most frequent fimbrial antigen.

    Summary: The administration of PGP (2.5 mg/kg/pig) by intraperitoneal (i.p.) route affected both erythrocytes and leukocytes by decreasing (P<0.05) or increasing (P<0.05) the number of these cells in the peripheral blood during the period of 3 weeks after the treatment. The percentage of lymphocytes was lowered (P<0.01;<0.05) in PGP-treated pigs farrowed to gilts or sows, respectively, on day 14 after farrowing when compared to that of the respective controls. On days 21 and 28 after farrowing the percentage of lymphocytes in PGP-treated pigs farrowed to gilts was significantly higher (P<0.05; <0.01, respectively) than that in control pigs. The cellularity and weight of the spleen in PGP-treated pigs from litters of gilts or sows were increased but differences were not significant as compared to those of nontreated controls.

    Summary: Immunoprophylaxis of neonatal diarrhea with allogeneic Igs resulted in about 50% decreased mortality among suckling pigs on a large swine farm. None of specific Igs-treated pigs developed the disease due to infection with Leptospira interrogans serogroup pomona and leptospires were not found in the tissues of those pigs. Conversely, leptospires were isolated from the blood and kidney cultures and other tissues (brain, mesenteric lymph nodes, liver) of nontreated pigs.

    Summary: Enterocyte receptors for F4 fimbriae of enterotoxigeneicEscherichia coli (ETEC) strains were detected in porcine feces byELISA using immobilized F4ac antigen and anti-brush border serum.Fourty of 44 pigs (91%) were classified identically as F4receptor-positive or negative by ELISA using fecal extracts orbrush borders. This finding clearly show that easily obtainablebody components (e.g., fecal samples and rectal swabs) can beused rather than brush borders to predict the state of resistance/susceptibility to F4+ ETEC infections, and that the test systemis potentially useful for identifying resistant pigs for breedingpurposes.

    Summary: Neonatal pigs (born to gilts or sows) were immediately after farrowing (from 0 to 6 hours after birth) perorally treated with allogeneic Ig (single dose of 200mg). A survival of Ig-treated pigs was better and resulted in the weaning of one more pig per litter as compared to the nontreated (control) pigs. The losses due to colibacillosis were much lower in the Ig-treated pigs (54 or 13%) than those recorded in the control pigs (96 or 25%). There was no difference in the rate of body weight gain between both groups of pigs during the observation period of three weeks.

    Summary: Both, peripheral blood lymphocytes (PBL) from peptidoglycanpolymer (PGP)-treated progeny of gilts or sows and spleniclymphocytes (SPL) from progeny of sows responded to ConAsignificantly higher on day 9th, 16th (P<0.01), and 21st(P<0.05), respectively, as compared to the responses of thecontrols. However, on day 28th PBL from PGP-treated pigs born tosows responded significantly lower (P<0.01) than those ofnontreated animals. Similarly, the responsiveness of SPL fromprogeny of gilts or sows was decreased (P<0.05; <0.01,respectively) on day 16th and 28th of age. PBL from both groupsof PGP-treated pigs were strongly stimulated (P<0.05; <0.01) withPHA on day 21st (progeny of gilts) and 28th (progeny of gilts orsows), manifesting a trend towards transformation to thelymphoblasts.

    Summary: BaypamunR-treated gilts had significantly higher (P<0.05) serum levels of total proteins (TP) and immunoglobulins (Igs) then nontreated gilts. The serum levels of Igs and TP in pigs born to gilt injected with BaypamunR were significantly higher 7 days (P<0.01) and 14 days (P<0.01; <0.05) after farrowing, respectively, than those in both control groups. Pigs born to BaypamunR-treated gilts survived much better (8.6/9.5 or 90.5% weaned pigs per litter) than did offspring from control sows (7.9/9.9 or 79.8%) during the preweaning period. No losses due to gastrointestinal syndrome (GIS) were observed in principals. Fifty percent (in litters of sows) or 83.3% (in litters of gilts) of total losses were diagnosed as GIS among control pigs.

    Summary: The ability of peptidoglycan polymer (PGP) isolated from Brevibacterium divaricatum to induce the proliferation of porcine peripheral blood lymphocyte (PBL) subpopulations was assessed. Crossbred (Yorkshire x Swedish Landrace x Dutch Landrace) pigs (1- to 5-week-old) born to gilts or sows were used as blood donors, and erythrocytes- and erythrocyte-antibody-complement rosettes were applied to determine changes in PBL subpopulation due to the PGP treatment. PGP (2.5 mg/kg) was given at Day 7 and Day 14 after farrowing by the intraperitoneal route, and rosette assays were performed at Day 9,14,16,21,28 and 35 of age. A trend toward increased T and B cell numbers and/or percentages was evident (P<0.05;<0.01) from Day 16 to Day 35 in PGP-treated pigs from multiparous sows or first-litter gilts. At Day 9 both lymphocyte subpopulations were decreased in PGP-treated pigs from litter of sows, but not from litters of gilts. These data indicate that PGP, if used as a purified agent, is a suitable modulator of nonspecific immunity in young pigs.

    Summary: The aim of our study was to determine whether or not the preparation of attenuated Parapox ovis, i.e. BPM is capable to enhance the proliferative responses of lymphocytes from jejunal lamina propria (LP), ileal mesenteric lymph node (MLN) and Peyer's patches (PP) of 4-week-old pigs orall inoculated with M1823 ETEC or non-ETEC strains expressing authentic or recombinant F4ac fimbrial antigen, respectively. The lymphocytes from MLN of M1823-treated pigs responded much stronger to BPM (P<0.01) and also to pokeweed mitogen (PWM; P<0.05) used as a positive control, than did the cells of noninfected pigs. Conversely, the cells from LP of the same pigs, although reacted better to PWM (P<0.05), were not stimulated with BPM. Strain 2407 activated lymphocytes from PP to respond better to BPM (P<0.05), but not to PWM stimulation. The cells from MLN of the same animals were stimulated much stronger (P<0.05) with both agents than did the control lymphocytes. The results indicate that BPM exhibited high in vitro stimulating activity (mostly for B cells from gut-associated organized lymphatic tissue, i.e. PP) as an immune response modifier , and thus, could be potentially synergistic with vaccinal antigen in stimulating the local (intestinal) immunity against enteric pathogens.

    Summary: The effects of new E. coli bacterin, Colimix that was used as a feed additive in the prevention of postweaning scours/setbacks were tested in 4-week-old pigs. Diarrhea was observed in 100% of controls, and in 84% of principals during the period of 3 weeks after first vaccination. None of the pricipals developed diarrhea after second vaccination. Eight (or 12.3%) of the 65 controls (and none of the pricipals) died from complications resulting from diarrhea. At 6 weeks of age pricipals were 10% heavier than the controls. A total serum IgG concentration of principals was significantly higher (P<0.05) than that of nonvaccinated controls.

    Summary: Diarrhea developed in F4-susceptible pigs in the control (1 of 5 pigs) and Colimix-treated/M1823-inoculated (4 of 7 pigs) groups, and 4 of the 4 affected principals died from complications resulting from diarrhea. None of the 8 F4-susceptible Colimix-treated noninoculated pigs developed diarrhea before 3 weeks of age. The F4-resistant pigs in the control and Colimix-treated/M1823-inoculated groups gained weight significantly faster from Day 6 to Day 21 after inoculation (P<0.05 and P<0.01, respectively) than did their F4-susceptible counterparts. There was no significant (P>0.5) difference in weight gain between susceptible and resistant Colimix-treated noninoculated pigs. The F4-susceptible pigs in the Colimix-treated group gained weight significantly faster from Day 6 to Day 21 after inoculation than their susceptible counterparts in the control (P<0.001 and P<0.01, respectively) and Colimix-treated/M1823-inoculated (P<0.001) groups.

    Summary: Both, non-ETEC strains 1466 and 2407, containing wild-type or recombinant plasmid, respectively, that encode expression of F4 fimbrial antigen induced a mild to moderate mononuclear/polymorphonuclear leukocyte (MNL/PMNL) infiltration in jejunal lamina propria of orally infected weaned pigs. Also, they neither caused destruction of villous epithelium nor swelling of the small intestinal mucosa. The use of F4+ ETEC strain M1823 was associated with marked lesions of jejunal/ileal villi, increased infiltration of MNL/PMNL and hyperplasia of Peyers patches. The results indicated that genetically-engineered F4+ LT- STa- STb- E. coli strain 2407 was able to invoke an infiltration of immunocompetent cells without inducing histological alterations in jejunal/ileal villous epithelium of experimentally infected pigs.

    Summary: Immunomodulation, as the term implies, can be used to designate either a suppression or an augmentation of an immune response. The necessity and capability of suppressing the function of the immune system are well recognized in such areas as organ transplantation and autoimmune disorders. However, in genaral, medically induced immunosuppression is not a practical concern in domestic food and companion animals. Conversely, augmentation of immunity has received much attention in domestic animal species and provides a means of increasing the host's resistence to disease. Various chemical and biological substances have been used and evaluated as immunomodulators in domestic food and companion animals. Other synonyms for immunomodulators that are frequently used include immunostimulators,immunopotentiators, imunotherapeutic agents, and immune response modifiers.

    Summary: Sixteen of the 53 principals, and 11 of the 50 controls tested were adhesion positive. Twelve of the principals and 1 of the controls developed diarrhea 1 or 2 days after the inoculation. The frequency of susceptible (26.2%) and resistant (73.8%) phenotypes was established on the basis of the presence or absence of the F4 receptors in 103 pigs as determined by the adhesion assay. During the 6-day to 3-week postinoculation period, pigs that were resistant to F4-mediated adhesion tended to have better rate of weight gain (P<0.05) than their susceptible counterparts.

    Summary: The objectives of the present study were to test the effect of crude peptidoglycan (monomer-PGM and polymer-PGP) preparations on PHA-induced responses of PBL and splenocytes from neonatal piglets farrowed to gilts or sows. PGM, when given (14mg/kg/i.p.) on Day 0 and 7 after farrowing suppressed the responses of PBL and splenocytes in piglets from litters of gilts (P<0.01; <0.05, respectively), and litters of sows (P<0.05;<0.05, respectively) as compared to the respective controls. The same amount of PGM when administered at Day 7 and 14 stimulated the responses of PBL and splenocytes in piglets from gilts (P<0.05), but did not induce significantly higher response of these cells in piglets from sows. PGP treatment (2.5 mg/kg/i.p.) on Day 7 and 14 after farrowing stimulated the PHA response of splenocytes in piglets from sows (P<0.01).     Summary: Anti-swine CD or SWC mAbs were used for immunohistochemical and cytofluorimetric characterization of lymphocytes from spleen, thymus, ileal mesenteric lymph node and lamina propria (LP) of jejunum/ileum of clinically normal 12-week-old pigs in order to determine normal values for that age groups.     Summary: Monoclonal antibodies (mAb) reactive with swine T-lymphocytes were used to characterize CD antigens of T cell subsets in gut-associated lymphoid tissues of weaned pigs. Lymphoid cells from lamina propria (LP) of 4-to 5-week-old pigs were identified as CD2a+ (0.3%), CD4a+ (1.83%), CD8a+ (22.13%), SWC1a+ (79.03%) and SWC2+ (4.03%) T lymphocytes. However, in an earlier study we found higher percentages of CD2+ (2.93%) and SWC1+ (97.27%) T cells in LP of 6-week-old pigs. It is likely that the numbers of at least these two T cell subsets are increasing in jejunal mucosa of pigs during the immunological maturation and acquisition of immunocompetence.

    Summary: Jejunal lamina propria (LP) of pigs inoculated with F4ac+ ETEC strain contained significantly higher number of CD2a+ (35,3%;P<0,05) and cytotoxic CD8a+ (35,3%; P<0,05) T lymphocytes when compared to the control noninoculated pigs (0,3% and 22,1%, respectively). In addition, a higher percentage of SWC1a+ T cell subset (68,6%; P<0,05) was determined in mesenteric lymph node (MLN) of inoculated pigs as compared to control pigs (48,2%). Finding of a smaller percentage of CD4a+ lymphocytes in LP of inoculated pigs (0,2%) was not significant (1,8% in the controls). It was concluded that immune response of T lymphocytes from MLN and LP could have a significant role in local cell-mediated immunity of weaned pigs diseased by colibacillosis.

This study demonstrated that both forms of F4 antigen, whether authentic (1466) or recombinant (2407), confer protective immunity against F4+ ETEC infection in postweaning pigs, and thus may be useful as an effective live oral vaccine.

    Summary: The aim of our study was to determine whether or not the preparation of attenuated Parapox ovis, i.e. BPM is capable to enhance the proliferative responses of lymphocytes from jejunal lamina propria (LP), ileal mesenteric lymph node (MLN) and Peyer's patches (PP) of 4-week-old pigs orall inoculated with M1823 ETEC or non-ETEC strains expressing authentic or recombinant F4ac fimbrial antigen, respectively. The lymphocytes from MLN of M1823-treated pigs responded much stronger to BPM (P<0.01) and also to pokeweed mitogen (PWM; P<0.05) used as a positive control, than did the cells of noninfected pigs. Conversely, the cells from LP of the same pigs, although reacted better to PWM (P<0.05), were not stimulated with BPM. Strain 2407 activated lymphocytes from PP to respond better to BPM (P<0.05), but not to PWM stimulation. The cells from MLN of the same animals were stimulated much stronger (P<0.05) with both agents than did the control lymphocytes. The results indicate that BPM exhibited high in vitro stimulating activity (mostly for B cells from gut-associated organized lymphatic tissue, i.e. PP) as an immune response modifier , and thus, could be potentially synergistic with vaccinal antigen in stimulating the local (intestinal) immunity against enteric pathogens.

Non-ETEC strain 2407 stimulated a strong proliferation of cytolitic CD8a+ cells in LP (P<0.01). pigs vaccinated with 2407 strain had more abbundant SWC1a+ and CD8a cells in MLN (p<0.05) or CD4a cells in PP (p<0.01), respectively. Numerous SWC1a cells were found in LP of villous epithelium, PP and paracortical/sinusoidal areas of MLN in M1823-treated pigs. In 2407-inoculated pigs marked infiltration/proliferation of CD8a+ cells were demonstrated in LP/PP and paracortex of MLN. Recombinant nontoxigenic strain 2407 seems to be a good source of protective (fimbrial) antigen for development of a safe and effective live oral vaccine against postweaning colibacillosis in swine.

    Summary: The effects of porcine non-enterotoxigenic E. coli (non-ETEC) strains 1466 and 2407 containing either wild-type or recombinant plasmids, encoding F4ac antigen were studied to establish their role in (1) causing mucosal damage of the gut, (2) inducing an inflammatory reaction of the gut-associated lymphoid tissue (GALT), i.e. in the small intestinal lamina propria (LP) and ileal mesenteric lymph node (MLN), and (3) stimulatig the protective immune responses mediated by CD+ T/B/leukocyte subsets in LP and MLN of experimentally infected weaned pigs. An increase of CD1+ and SWC2+ cells was detected by single colour FCM among B and T lymphocytes, respectively, from LP and MLN of 2407-infected pigs. A strong proliferation of CD45+ cells was observed in LP/MLN of pigs infected with either 2407 or M1823 strain. The later strain activated CD8a+ cytolytic T cells in these tissues. The use of non-ETEC strain 2407 was associated with a marked proliferation of CD1+ and SWC2+ cells in both tested compartments of porcine GALT.

The IPP and JLP of these pigs contained 2 to 25 times more of CD4a+ T cells than those of 4- of 5-week-old pigs. However, the proportion of CD4a+ and CD8a+ cellswas much higher (2-15 times) in PB and T of 4-week-old pigs. Species-specific SWC1/SWC1a T cells were more frequent (2-fold to 6-fold) in MLN, PB, S, BM, and IPP of 4-week-old pigs compared to 8-week-old pigs, but roughly equal in T and JLP of 4- or 5- and 8-week-old pigs, respectively.     Summary: Postweaning colibacillosis is an economically significant disease in swine for which no effective vaccine is available. Porcine F4+ enterotoxigenic Escherichia coli (ETEC) strains are recognized as a significant cause of diarrheal disease in suckling and weaned pigs. It is assumed that the harmful effects are due entirely to enterotoxins and that F4+ nontoxigenic (non-ETEC) strains of E.coli could be an effective and safe source of fimbrial antigen for the development of live oral vaccines against postweaning colibacillosis mediated by F4+ ETEC strains. The current study have investigated the harmful effects and immunogenicity of F4ac+ ETEC strain (M1823) as well as of recombinant and authentic non-ETEC strains (2407 and 1466, respectively) in 3-week-old pigs in order to establish the role of F4 antigen in: (1) producing diarrhea and mucosal damage in the small intestine, (2) inducing an inflammatory response of gut-associated lymphoid tissue (GALT), and (3) stimulating local immune response of the GALT mediated by the cellular elements in jejunal/ileal lamina propria (LP), Peyers patches (PP), and ileal mesenteric lymph node (MLN). Non-ETEC strain 2407 expressing F4ac+ antigen induced only a mild mucosal damage and a moderate leukocyte infiltration in jejunum and ileum of inoculated pigs as compared to the findings in noninoculated or those animals that were infected with F4ac+ ETEC strain M1823. Among lymphocytes from LP of M1823-, 1466-, or 2407-inoculated pigs CD2a+ and CD8a+ cells were predominant (P<0.05), as compared to those found in control pigs. The number of CD4a+ cells was significantly (P<0.05) higher in PP of pigs inoculated with non-ETEC strain 2407 than that in noninoculated pigs. Also, in MLN of 2407-inoculated pigs CD8a+ and SWC1a+ cells were more abundant (P<0.05) than those in the controls. F4ac+ non-ETEC strain 2407 and, to a lesser extent 1466, activated lymphocytes from PP and MLN to respond better to common mitogens (PHA, PWM, LPS), purified fimbrial (F4ac) antigen, or immunologic response modifiers (IRM; BaypamunR, peptidoglycan monomer). The results indicate that F4ac+ non-ETEC strain 2407 exhibited high immunogenicity and, thus, could be the best candidate for the development of safe and effective live oral vaccine against postweaning diarrheal disease caused by F4+ETEC strains.
