Summary: The effect of freezing on the infectivity of Toxoplasma
gondii tissue cysts was analysed. Tissue cysts of two different isolates
(TG no.3 from Zagreb and NT from China) in muscles and brains of
experimentally infected pigs, with or without infected murine brains, were
stored at -7, -12 or +4 C during 4 days. Then, the tissue samples were
homogenized by pepsin HCL digestion and subcutaneously injected into SPF
mice. Six weeks later the mice were sacrificed by euthanasia and their
brain tissue microscopically examined for tissue cysts. Toxoplasms were
found in no mice injected with previously frozen samples, whereas in those
injected with control samples, tissue cysts were demonstrated.

Summary: The effects of 95% and 75% ethanol, 100% methanol and 10%
formalin on the viability of Toxoplasma gondii sporulated oocysts were
studied. The oocysts were treated at room temperature for 1, 2, 4 and 24
hours respectively. Then they were washed and subsequently subcutaneously
injected into SPF mice. Parallely, non-treated oocysts were bioassayed as
controls. Six weeks after injections the mice were euthanatized and their
brains microscopically examined for T.gondii tissue cysts. The results with
the treated oocysts were comparable to those of the non-treated controls.
Thus, the tested alcohols and formalin failed to kill T.gondii oocysts.

Summary: Toxoplasma gondii oocysts survived a 24-hour treatment with
1% and 2% water solution of Virkon S (potassium peroxisulphate) and for at
least seven weeks when dried and stored at room temperature and with a
relative humidity of between 30 and 68%, but were inactive after three
weeks at -20 C, and after 15 minutes at 58 C. Tissue cysts survived heating
at 50 C for 15 and 30 minutes, but were inactive at 58 C after 15 minutes.
The viability of oocysts and tissue cysts was assesed by bioassays on mice.

Summary: Tissue cysts of Toxoplasma gondii in pork meat died when
frozen at -7 C during 4 days and after heating at 58 C during 15 minutes.
The oocysts from cat faeces also died when heated at 58 C during 15
minutes, but when frozen at -20 C they died after 3 weeks only. When dried
and stored at room temperature, with 30-68% of relative humidity, they
survived at least 7 weeks. Furthermore, they showed high resistance to
disinfectans Aldesol, tincture of Hibisept, Izosan-G and Virkon S, and to
concentrated alcohols ethanol and methanol as well as to 10% formalin. The
viability of the treated tissue cysts and oocysts was bioassayed on mice.

Summary: Within the study of the use of irradiation to control
infectivity of food-borne parasites we have studied the effect of X rays or
gamma rays on the infectivity of T.gondii tissue cysts. The results were
assesed by bioassays on cats and/or mice. It was found that a complete
inactivation was achieved with doses of 0,4 to 0,7 kGy, depending on the
isolate. In order to study the possibility of killing sporulated oocysts
with chemicals we have examined the effect of 4 currently used
disinfectants (Aldesol, Hibisept Tincture, Izosan-G and Virkon S), and of
the concentrated ethanol, methanol and 10% formalin. The chemicals have
acted at room temperature and the results were assessed by bioassays. None
of the tested chemicals proved efficent in killing the oocysts.
