    Summary: In last several years a veriety of instrumental methods usingmass spectrometry have been developed, to determine drugs andtheir metabolites in biological sample. Application of theGC/MS/COMP system in qualitative and quantitative determinationsof different classes of drugs and metabolites has spreadprimarily to biochemistry, medicine and toxicology in addition tocontrol the use of doping agents in sport. The methods of samplepreparation and derivatization reagents used are suitable forGC/MS analysis of parent drugs and metabolites with respect ofvolatility thermal stability and fragmentation pattern. Asexamples determinations of metabolites in urine samples afteradministration of mefenorex, etafedrine, and drug Mucaine arepresented. In addition, influence of different factors,particularly those inhibiting metabolism of doping agents arediscussed.

    Summary: Use and misuse of anabolic steroids in sport is reviewed together with the presentation of the methods and procedures used for unambiguous determination and structural confirmation of endogenous as well as exogenous steroid hormones. Examples are presented from the experience of the doping control performed in the USA, and the discussion and conclusions are based on the policy of the IOC Medical Commission and of the International Federation of Clinical Chemistry. Side effects of administration of anabolic steroids as well as analztical approach of their determination on biological material is discussed.

    Summary: Physico-chemical properties (lipophilicity-log P and molecularmass-M.m.) and biological activity (norepinephrine inhibition-logA) of phenylalkylamines were quantitatively analyzed. Topologicalindices used for correlation analysis include Wiener's number,Randić connectivity index, Balaban's index andinformation-theoretic index. The comparasion of four topologicalindices shows that Wiener's number is the most convenient as thestructural parameter in quantitative structure-propertyrelationship (QSPR), as well as in quantitativestructure-activity relationship (QSAR) analysis ofphenylalkylamines. A number of indices, except Randićconnectivity index, are found to be equieffective or superior tolog P in correlations with biological activity.

    Summary: Using a set of 15 quinolone derivatives analysed by means of 9chromatographic systems, two different decision making tools inHPLC were tested. Information content and numerical taxonomy wereapplied to select the optimal set of rationally chosenchromatographic systems. Selectivity was evaluated on the basisof retention data of individual substances in all of the systems.Standardized k' data were shown to be more relevant thanretention times or unprocessed k'. When used with suitable inputdata, both methods proved to be powerful tools in selecting theoptimum combination of HPLC systems to analyze a large number ofreal substances having complex structures.

    Summary: Metabolic and binding properties to plasma proteins of steroidhormons taking testosterone as an example compound are presented.Using gel filtration method the binding parameters oftestosterome were determined. One set of equivalent binding sitesof low affinity (K=17400 L/mol) for binding of testosterone toHSA was found. The percent of binding was from 12% to 30 %depending on the concentration of the ligand. Binding curvesindicate nonsaturable process.

    Summary: Metabolism of testosterone and formation of the end products of its metabolism by catalytic activity of cytochrome P-450 enzymes is presented. Selected reactions of testosterone metabolism and influence of other drugs, including steroids are discussed in relation to the formation and excretion of metabolites. Results of clinical studz are presented (the clinical studz was performed at the University of California, Los Angeles, Department of Pharmacology, Dr. Don Catlin). On the basis of the GC-MS analysis the structure of metabolite was determined in the urine (11beta-hydroxytestosterone) as a new urinary metabolite and the ratios of hydroxylated metabolites was proposed as a paramtere indicating administration of egsogenous testosterone.

    Summary: Catalytic properties of human cytochrome P450 (CYP) enzymes are reviewed regarding their involvement in metabolism of both the clinically used drugs and the drugs that are banned or restricted in sport. Of about 36 families of CYP enzymes identified until now, only few of them (familiesCYP 1, 2 i 3) are involved in metabolism of drugs in humans. Similarly to other drugs, also metabolism of doping agents by CYP enzymes is influenced by a number of factors including genetic polymorphism. Clinically important drugs are presented in tables regarding their participation as substrates, inductors or inhibitors of cytochrome P450 enzymes in humans. These data are proposed for use as information for prediction of drug-drug interactions and toxic effects of drugs and certain chemicals.

Displacement and the mechanisms of binding was investigated in the presence of other anabolic steroids (stanozolol).     Summary: A sructure-toxicity study of nitro compounds, which serve as organic explosives and propellants, is caried out by topological indices. Toxicity data are taken from Rosenblatt et al. We considered bioconcentration factor in fish and beef, and acute toxicities to rodents. Stucture-toxicity models with several kinds of indices are tested. The simplest and best models are obtained by using connectivity indices. In the present theoretical framework these are the models with highest predictivity for considered biological properties.

    Summary: In the present work we investigate structural requirements of cimetidine as ambidentate ligand for binding to CYP enzymes, horseradish, Ac-MP-8 as well as with a model porphyrins Co(II)PPIX and Co(II)TPP.     Summary: The compounds analyzed in this report are 17 new derivatives of 3-(phthalimidoalkyl)-pyrazolin-5-ones recently synthesized in our group. All studied compounds were tested for their mitodepressive activity upon the growth of Lepidium sativum L. seeds. The inhibiton of the seeds growth was significant for derivatives of glycine and of alanine, which are used in QSAR study of supstituted pyrazoles with known cytostatic activity (ED 50) on the HeLa cells. Topological indices found to be equieffective or superior to log P in correlations with biological activity.

    Summary: The lipophilic character of steroids has an important role in regulating the biological activity of these compounds.Lipophilicity and physico-chemical properties of steroid hormones were quantitatively analyzed.In this purpose, partition coefficient, log P,has been calculated according to the Rekker method and for correlation analysis the topological indices were used.The best models have been obtained by using Wiener number and Balaban index modified for heteroatoms.

    Summary: Zizyphus jujuba Mill. is a small tree, which grows in China,Australia, America, Asia and Mediterranean.This plant is mostly used in folk medicine to heal bronchitis, diarrhea and ulcers.Chemical investigation of this plant indicated the presence of flavonoids and phenolic acids. Experiments were carried out by TLC methods in thirteen systems and selection of the most efficient TLC-systems were carried out by the methods of numerical taxonomy.

    Summary: Seven flavonoids have been isolated from the leaves of Paliurus spina-christi Mill. by the chromatographic methods. Their structures have been established by the spectroscopic methods (UV,NMR). The isolated flavonoids were characterized as derivatives of quercetin, kaempferol and scoparin.Derivatives of quercetin have exhibited an antimicrobial effect.The efficiency of ten TLC-systems for separating flavonoids was compared by using the information theory and numerical taxonomy.

    Summary: Binding of (+)-, (-)-isomer and racemate of etomoxir to the humanserum albumin (HSA) was studied using gel filtration method. Theexperimental results showed statistically significant differencein affinity of binding of (+)-isomer when compared tothat for (-)-isomer and racemate (p<0,05).The binding wasinfluenced by the presence of competitive ligands stearic acidand stanozolol.The binding kinetics of the (+)-isomer wasdifferent compared to (-)-isomer and racemate also in thepresence of the competitive ligand anabolic hormone stanozolol.It was concluded that stanozolol affects binding of the(+)-isomer predominately at its high affinity sites.
