    Summary: Mustard derivatives of ethyl-choline and hemicholinium-3 havebeen suggested as possible specific cholinergic neurotoxins. Astructural analog of hemicholinium-3 (toxin 7) was added tosynaptosomes prepared from the cortex, striatum or hippocampus ofrat brain. HACU was significantly decreased by addition of toxin7, while GABA or dopamine was not changed. Incubation of corticalsynaptosomes with the GM1 prevented the decrease in HACU. Twonewly synthesized hemicholinium-3 analogs (toxin 5 and 6) causeda larger decrease in HACU then toxin 7 and ethyl-cholineaziridinium.

    Summary: Pyruvic acid and acetaldehyde react with substitutednitrosobenzenes to give the corresponding N-phenylacetohydroxamicacids. A mechanism for these reactions involving three sequentialsteps is proposed. The first step is nucleophilic attack of thenitroso group on the carbonyl group, the second step is protontransfer, and the last one is a decarboxylation.

    Summary: L-Dopa and ŕ-methyldopa were attached by an ester linkage toŕ,á-poly(N-hydroxyethyl)-DL-aspartamide (PHEA) by means of1-benzotriazolylcarbonyl group. Release of the drugs based onhydrolysis of PHEA adducts was studied in vitro. Characterizationof the PHEA-L-dopa adduct was carried out in vivo using anintracerebral microdialysis technique in order to evaluate theprolonged release eventually achieved. A new method forpreparation of several simple esters of L-dopa and ŕ-methyldopais described.

    Summary: QSAR models are characterised by two assumptions with respect tothe relationship between the chemical structure and thebiological activity of a compound. The first is that one canderive quantitative measure from the structure of those globaland local properties of significance to the biological activityof a compound. The other assumption is that one canmathematically describe the relationship between biologicalproperty one wishes to optimize and the molecular propertiescalculated from the structure. The most popular QSAR model is thelinear free energy, extrathermodynamic or Hanschs model. In ourtimes QSAR became a standard tool in drug design.

    Summary: A series of new halo-derivatives of pyridoxine- andpyridoxal-3-O- or 5-O-phosphates have been synthetized. In thereaction of partially protected pyridoxine and pyridoxal withphosphorusoxychloride dichlorophosphates were obtained. Thesecompounds reacted with NaF under catalytic action of18-crown-6-ether, giving the corresponding difluorophosphates.The direct fluorination of pyridoxal-5-phospate withdinitrofluorobenzene yielded pyridoxal-5-O-monofluorophosphate.

    Summary: The synthesis of pyridoxal-5-fluorophosphate (PLPF) has beenachieved starting from dinitrofluorobenzene and pyridoxalphosphate (PLP) protected in the aldehyde region. PLFP as a PLPanalogue is an ideal probe for structural studies ofcoenzyme-apoenzyme liganding, especially so in the investigationof the phosphate binding region of the protein molecule. Thereconstituted AST-PLFP enzyme shows a lower catalytic activityand has a more tightly bound prostetic group in comparison withthe natural holoenzyme.

    Summary: Liposomes with clindamycin hydrochloride were prepared usingeither soya lecithin and cholesterol or hostaphate andcholesterol. In vitro dissolution studies showed sustainedrelease of the drug from hostphate liposomes compared to lechitinliposomes. Clinical treatment of Acne vulgaris with a lotion ofliposomal drug shows better efficacy than non-liposome lotionforms (esspecially of the treatment of pustules where clinicalimprovement was 77 % of initial number).

    Summary: The total drug content of microspheres was dependent on thepartition coefficient and an increase in the partitioncoefficient caused a decrease in the drug content of finishedmicrospheres due to drug migration to the outer organic phase.The ensemble drug release from microspheres was followed by thepaddle method and by potentiometric titration with a pH-stat.Spherical Higuchi equation, especially biphasic first orderkinetics, gave the best fit for the release.

    Summary: Results of these studies strongly suggest that coencapsulation oftwo different neuroprotective factors into biodegradable polymersmay be used as a novel mean for delivery of these agents into theCNS. It was also demonstrated that genetically modifiedfibroblasts can be encapsulated into biocompatible, biodegradablespheres retaining their viability and capacity to continouslysecrete nerve growth factors for at least two months. Nervegrowth factor (NGF) and monosialoganglioside (GM1) weremicroencapsulated in copolymer of lactic and glycolic acid,tested in vitro and in selective experiments in vivo in animalswith unilateral cortical devascularizing lesions.

    Summary: A new method for preparation of non-steroidal antiinflammatorydrug esters and ŕ-methyldopa esters by means of 1-benzotriazolecarboxilic acid chloride (BtcCl) was described. BtcCl reactedwith the carboxylic functional group in the drugs formingreactive benzotriazolides. Benzotriazolides with hydroxycompounds gave the corresponding esters. In the case ofŕ-methyldopa Btc served as an N-protecting and C-activatinggroup. In the reactions of esterification simple alcohols andŕ,á-poly(2-hydroxyethyl)-D,L-aspartamide were used. Release ofactive substances from polymer-drug conjugates was studied invitro.
