Summary: The effect of newly synthesized derivative 5-aryl-5-hydroxy
-2-/aryl-(hydroxyimino/ethyl/-1-cyclohexanone, on growth and
zearalenone accumulation by toxigenic mold Fusarium graminearum
ZMPBF 1244 was studied in yeast extract-sucrose (YES) medium to
determine the possible use of this compound as a mean of
controlling zearalenone accumulation.Experiments were carried out
in a stationary culture at temperatures of 10ř and 20řC , respec-
tively, with and without "temperature shock" for 25 days.Mycelial
dry weights were determined gravimetrically, and concentration
of zearalenone (ZEA) were measured fluorodensitometrically using
a Camag TLC Scanner.In experiments with a "temperature shock" and
at pH 5,5 concentration of investigated derivative higher than
15,0 Šg/mL of liquid medium completely inhibited growth and ZEA
accumulation after 15 days of incubation at temperatures of 10ř
and 20řC.Concentrations of 5,0 Šg/mL and 10,0 Šg/mL partially
inhibited growth and decreased ZEA accumulation by 45% and 70%
respectively.Inhibition was depending of the time of cultivation
and pH values of substrate.

Summary: The relationship between mold biomass and the biosynthesis
of
aflatoxins B1 and G1 (AFB1 and AFG1) on solid substrates (whole
and crushed wheat grain) at temperatures from 15-35řC and a water
content in the substrate of 20-40% has been investigated. The
experiments have been carried out with the aflatoxigenous mold
Aspergillus parasiticus NRRL 2999 in pure culture and in mixed
culture respectively, the latter with the mold Fusarium
graminearum ZMBF 1244. Mold F.graminearum does not produce
aflatoxins and chromatographically similar compounds.
The amount of biomass was estimated by measuring the chitin
content, and the aflatoxin concentration by means of a
fluorodensitometer. It has been established that the biosynthesis
of the examined aflatoxins and their ratio primarily depend on
the temperature of cultivation, rather than on the growth of the
mycelium.The biomass of the mixed culture of A.parasiticus and
F.graminearum after six weeks of cultivation reduces the amount
of AFB1 by 45-60%, and the amount of AFG1 by 35-70%. The
decrease of both toxins concentration is more pronounced in the
substrate with a higher initial water content and a higher
temperature of cultivation.

Concentrations of DHA and DHT of 0,1 and 0,2 mmol/L respectively
stimulated, but concentrations higher than 0,5 mmol/L of these compounds,
mold growth and ochratoxin A synthesis partially inhibited. In experiments
with 0,2 mmol/L of BrDHT mold growth and synthesisof ochratoxin A were
inhibited completely depending on time and temperature of cultivation.

and the amount of aflatoxin G1 by 25-60% in respect to highest values
obtained in the process of biosynthesis.The decrease of concentration of
both toxins is more pronounced in the substrate at the higher initial water
contents and at the higher temperatures of cultivation.

Summary: The effect of dehydracetic acid (DHA) and its newly
sinthesized
analogues,4-hydroxy-3-(p-toluoyl)-6-(p-tolyl)-2H-pyrane-2-one
(DHT) and 5-bromo-4-hydroxy-3-(p-toluoyl)-6-(p-tolyl)-2Hpyrane-
2-one (BrDHT), were tested for antifunal and antiaflatoxigenic
activity in experiments with the moulds Aspergillus parasiticus
NRRL 2999 and Aspergillus ochraceus NRRL 3174 in the yeast-extract-sucrose
(YES) medium.Investigations were carried out to determine the possible use
of these compounds as a means of controlling aflatoxin and ochratoxin A
accumulation. Experiments were carried out in a stacionary culture at
different temperatures during 15 and 28 days. The investigated compound DHA
and DHT are poor inhibitors. Very low DHA and DHT concentrations
even stimulated mould growth and production of this toxins. In experiments
with BrDHT analognes mould growth and synthesis of aflatoxins and
ochratoxin A were inhibited completely depending on the time and
temperature of cultivation. In all experiments mycelial dry weights were
determined gravimetrically. Concentration of this toxins were measured
fluoro-
densitometrically, and qualitative confirmation were made with mass
spectra.

but concentrations higher than 5,0 ŠM/L produced an inhibitory effect.In
the presence of low BrDHT concentrations mold growth
decreased by 60% and 80% and toxin concentrations by 85% and 100%,
respectively.

Summary: The effect of newly synthetized Schiff base on the growth
and biosynthesis of zearalenone by mould Fusarium graminearum ZMPBF
1244 was studied on barleycorn to determine the possible use of this
compound as a means of controlling zearalenone accumulation.
Experiments were carried out in a stationary culture at tempera- tures of
10,15 and 20řC, respectively, with and without "temperature shock" for 42
days. In experiments with a "temperature shock", Schiff base inhibited the
growth of mold by 70% and zearalenone accumulation by 95% in respect to
highest values obtained in control. Using different concentrations of
Schiff base we found that, although Schiff base did strongly inhibit
zearalenone accumulation by 95%, it did not completely prevent its 20,0
Šg/g of barleycorn.

Summary: The larvicidal effect of concentration of aflatoxin B1 and
diacetoxyscirpenol at temperatures of 20 and 35řC, on the
larvae Artemia salina was investigated.Concentrations resulting
in 50% mortality, determined with these organisms exposed to
known mycotoxin for 24 hours were(Šg/mL): aflatoxin B1 1.0, 2.0
and 3.0, and diacetoxyscirpenol 0.8, 1.8, 2.0 and 4.0.In a study
of aflatoxin B1 and diacetoxyscirpenol synergism in acute toxicity, the
LD50 values of toxin pairs exhibited a distinct increase in toxicity.
