Summary: The investigations (using immunologic, electrophoretic, chromatographic and sedimentation, thermodynamic and kinetic methods) focus on: 1. stress induced biochemical changes (such as concentrations of NO synthetase, Zn, Cu and Cr ions, lysosomal enzymes, nuclear glycoproteins, lectines, adenylate cycle components and other stress proteins) in animal model of the induced acute and chronic stress, in sera of Vukovar prisoners, and in trophoblasts after ultrasound treatment. 2. The synthesis of newly designed ALP inhibitors in order to kinetically distinguish between tissue specific isoenzyme what is of great diagnostic importance in clinical chemistry. 3. Functional and kinetic study of ecto-ADPase and ATPase activity of kidney brush border membranes. 4. Study of the role of CFTR protein in the regulation of chloride transport across plasma membranes. 5. Study of the active site of cyclophilin as binding protein for the immunosupressor cyclosporin A widely used during organ transplantation. 6. Diagnostic significance of N-acetyl-beta-D-glucosaminidase and alaninaminopeptidase in monitoring transplantation of Langerhans cells in rats with induced diabetes. 7. Effect of halogenated pyridoxal phosphate (Cl, F in phosphate) on the activity of aspartate aminotransferase in order to define the functional role of the phosphate binding region in the catalytic reaction. Biological effect of the fluorinated prostetic group. 8. Biochemical changes revealing endemic nephropathy in ochratoxin animal model of the disease.

Research goals: 1. Study of the protein structure-function relationship, of their synthesis and localization and of the mechanism of their interactions with specific ligands. 2. Determination of biochemical changes induced by: chronic and acute stress in animal models, prenatal ultrasound treatment, and in prisoners from Vukovar. Effect of ageing on biochemical stress response. 3. Defining diagnostic relevance of changes in catalytic concentrations of enzymes which may improve early diagnosis of imairments due to transplantation, cystic fibrosis, mucopolysacharidoses, diabetes, endemic nephropathy, oncogenesis, stress and ageing. Better understanding of the molecular basis of listed diseases. 4. Design of analytical and preparative methods for glycoprotein research. 5. Introduce and standardize novel diagnostic methods. 6. Define analytical and biological drug interferences with the results of clinical laboratories. 7. To investigate and help in dealing with health problems specific for Croatia.
