Summary: MOET investigations (multiple ovulation end
embriotransfer) were performed in more then 60 cows and heifers. Induction
of superovulation was investigated using different hormone preparations
(Folligon, Pregmanon, Folltropin, F.S.H.-P), sinchronization of estrus
cycle (Anifertil), and the results of induced superovulation were followed
up by ginecological (rectal and vaginal) and ultrasound examination. A 5
MHz transducer and a high-quality scanner were used for transrectal
evaluation of the reproductive organs, primarly ovaries during a
superstimulation regimens and superovulatory response (corpora lutea,
follicles, cystic ovarian conditions). That way we could determine the
number of corpora lutea and predict the recovery rates of embryos. Embryos
were collected using non-surgical flushing (gravitation and aspiration)
with PBS (Dulbeccos phosphate buffered saline) medium, and then evaluated
and classified using IETS (International Embryo Transfer Society) criteria.
One part of embryos were immediately transferred in sinchronized recipients
and rest were cryopresevated using special mediums (glycerin) and plunged
into liquid nitrogen (-196 degrees C). When we succeded to get new
recipients, embryos were thawed by "one step" procedure, evaluated in
accordance to IETS and transferred.
Research goals: The goals of this project in the first year were:
investigationson on the possibilities for inducing superovulation,
selection of the most reliable procedure, inspection and estimation of
superovulatory responces by ultrasonography and, finally, flushing and
transfer of collected embryos in recipients. Mentioned aims of this
project for the first year were completely achieved, and besides we
accomplished cryopreservation and thawing, and finally transfer of those
embryos in three recipients. Further researches will be directed on the
collection of the embryos from at least 50 donor cows, and transfer to at
least 25 to 30 recipients. The collected embryos will be cryopreservated
with a wiew to preserve and later use of possible overplus, untill the new
recipients would be prepared. Besides, this procedure will be used to
perform embryo transfer in as possible as more recipients, so in the
beginning of the second year and during the third year of researche, we
will have a specific comprehending in the efficiency of the
cryopreservation/thawing procedures. This researche will be aimed on the
improvement and simplification of the procedures used for freezing and
thawing of bovine embryos. In that period our investigations will be
directed on evaluation and categorization of collected embryos in
accordance with international standards, and on the possibilities in
improving the mediums for flushing, maturation, freezing and thawing of
bovine embryos. Our investigations will be completed with performing a
selection of the top cows of various breeds (40 to 50 cows), to form
nucleus herds for quality embryos supply. In the contining of the
project, investigations will be directed to maturation and fertilization
of bovine oocytes in vitro.
