Summary: The goal of the investigations proposed, is the development
of the following new types of potentiometric sensors (ion-selective
electrodes) and biosensors: surfactant, calcium, magnesium, halogenide,
enzyme sensor. The investigations should comprise the synthesis of sensor
material, selection of sensor type and conducting substrate, selection of
plasticizer and solvent mediator, response of the sensor towards the
species investigated, analytical and physicochemical sensor
characterization, application of the sensors for the quantification of
species investigated, using direct potentiometry and potentiometric
titration method. Biosensors (enzyme electrodes) should be characterized
according to the following aspects: construction of enzyme electrode,
measuring of actual enzyme activity, influence of the pH value and ionic
strenght of the medium on enzyme activity, analytical characterization,
the application possibilities for the following of enzyme reaction
kinetic, investigation of the influence of inhibitors and activators etc.
Research goals: The main goals of proposed investigations are:
-development of the rapid, simple, accurate and inexpensive analytical
methods for routine determination of species investigated, in pure and
complex materials -the overcoming of the disadvantages of the existing
routine analytical methods (visual titrimetric methods, spectrophotometric
methods etc.) for actual determination: visual detection of the end-point,
time-consuming and labour intensive methods, interferences at the
end-point location by the coloured and turbid samples, use of toxic
organic solvents, etc. -the overcoming of the limitations of up to now
developed ion-selective electrodes and biosensors: lack of selectivity and
sensitivity, problems connected with conditioning of electrodes,poor
mechanical properties by some types of electrodes, increasing of life-time
of electrodes, etc. -reducing the analyte amount. The results of
investigations should contribute to: -better understanding of the
phenomenon on the ion/organic molecul-sensor interface, -better
understanding of the reaction-mechanism inside the sensor's (ion-selective
electrode, biosensor) membrane, -analysis (determination, monitoring) of
specific chemical and biological/biochemical species, -introduction of new
chemical/instrumental analysis of materials, -development and
commercialization of chemical sensors and biosensors.
