Summary: The biological effect of respirable asbestos fibers,
prepared for animal model experiment as well as inert polystirene
microspheres on a free lung cell population obtained by bronchoalveolar
lavage of intrabronchial instilled experimental animals, will be
examined. We anticipate that the results will demonstrate in what way the
instilled agents influence the functional ability of phagocytes, and also
provide insight of their redistribution among the cells. The presumed
development of lung fibrosis in experimental animals will be demonstrated
by standard histological methods. The correlation between the
concentration of the forementioned agents and the length of exposure with
the results of the examined phagocyte functional ability will be
determined by using adequate biostatistical methods.
Research goals: The aim of the study is to examine the functional
ability of lung and alveolar phagocytes exposed to asbestos fibers and
inert particles "in vivo". The investigation of the functional ability
of phagocytes under the controlled experimental conditions will contribute
to better understanding of the role of the phagocytes in the transport,
deposition, clearance, retention and redistribution of phagocyted
particles, thereby throwing significant light on the mechanical-irritative
and physical-chemical theory on the occurrence of mineral dust related
diseases. By means of adequate biostatistic tests, important parameters of
early detectable biological effects of the applied agents would be
established. The primary aim of this study is to provide a significant
scientific contribution to the unclear problem of etiology and
pathogenesis of lung fibrosis caused by fibrous mineral dusts, as well as
in prevention of disability caused by development of occupational
pneumoconiosis.
