Summary: The effect of endocrine pancreas cells transplantation on
the activities of the key enzymes in glycolytic, gluconeogenic and
pentose phosphate pathway in the liver of alloxan diabetic rats and mice
are investigates, especially with regard to the duration of disease.The
results are compared with the effect of insulin administration following
several week. In the both groups of diabetic animals (that received
transplantat or treated with insulin) the hepatic enzyme activities are
compared with activities in healthy animals (healthy control) and in
diabetic animals immediatly before the therapy is started (pre-therapy
diabetic control), as well as in non-treated diabetic animals with the
same duration of disease as therapied animals (parallel diabetic
control). In the second part of our work we have investigated whether
iron deficiency in postnatal anemia of pigs is associated with the
changes in glucose metabolism. Namely, low transplacental exchange of
iron, rapid neonatal growth, and diet inadequacy cause iron deficiency in
baby pigs, which is economically impotrant disease in large pig
production units. We have examined parallely hematological parameters,
serum iron level, and the concentration of glucose and its precursors in
the blood. In addition, the activity of some regulatory enzymes in
hepatic glucose metabolism are measured. The results are compared with
the results obtined on healthy pigs (iron supplemented group).
Research goals: The main goal the first part of investigation is to
establish in which extend, and how long, hepatic carbohydrate and lipid
metabolism is corrected after endocrine pancreas cells transplantation in
comparison with insulin therapy. Additionally,we can get an information
about the time in which the transplantation give the best results. In the
second part of investigations, the parallel determinations of
hematological parameters with the concentration of glucose and some
regulatory gluconeogenic substrates (lactate,pyruvate) in the blood, and
also with hepatic enzyme activites, could demonstrate whether increased
glucose dependance exist in iron-deficient postnatal anemia of pigs.
Additionaly, the results can contribute to our understanding whether
hypoglicaemia of neonatal pigs is associated with increased glucose
metabolism induced by iron deficiency.
